吲哚美辛诱导宫颈癌Hela细胞凋亡机制及其联合顺铂的协同作用研究

摘　　要：目的研究吲哚美辛诱导Hela细胞凋亡作用机制及其联合顺铂对Hela细胞生长抑制的协同作用。方法采用MTT法检测吲哚美辛、顺铂及两者联合应用对Hela细胞的增殖抑制作用,Western Blot法分析凋亡相关蛋白的表达情况。结果吲哚美辛与顺铂联合用药比单独用药具有更强的抑制效果(P<0.05)。与空白对照组比较,给药组Bcl-2、Procaspase-9、Procaspase-3的表达显著降低(P<0.05)。结论吲哚美辛可协同顺铂增强对Hela细胞的抑制作用。吲哚美辛诱导宫颈癌Hela细胞凋亡的机制可能涉及线粒体凋亡通路的激活。OBJECTIVE To investigate the meachanism of indometacin on apoptosis of Hela and the Synergistic suppressive effect of indometacin and cisplatinum on Hela.METHODS The inhibitory rate of indometacin,cisplatin and indometacin combined with cisplatin on Hela was tested by MTT assay.The protein expression on apoptosis was detected by Western Blot.RESULTS Indometacin combined with cisplatin have a stronger effect than monotherapy.Compared with DDP equal dose group,the expression of Bcl-2,Procaspase-9 and Procaspase-3 in the indometacin group is significantly lower.CONCLUSION Indometacin has synergistic effect with cisplatin on the suppressive effect of Hela.The meachanism of indometacin on apoptosis of Cervical Cancer Cell Hela is probably involved in the activation of mitochondrial apoptotic pathways.
