摘 要:目的研究黄芩汤治疗克罗恩病(CD)的分子作用机制。方法检索TCMSP数据库获得黄芩汤潜在活性成分及靶基因;GEO芯片、Genecard及OMIM数据库收集克罗恩病疾病靶基因,两者取交集得到药物-疾病靶基因,并经过Cytoscape软件将结果可视化,通过网络拓扑算法筛出关键靶基因。利用Bioconductor中R包clusterprofile对交集靶点进行GO生物进程富集和KEGG通路分析,结合文献分析黄芩汤治疗克罗恩病的分子作用机制。结果得到黄芩汤治疗CD关键靶点65个;槲皮苷、山柰酚、柚皮素、汉黄芩素等可能是黄芩汤治疗CD的主要活性成分;作用于ESR1、AKT1、IL6等靶点,参与PI3K-Akt信号、HIF-1信号、IL-17信号、TNF信号等通路发挥治疗CD作用。结论黄芩汤通过多组分-多靶点-多通路治疗CD,同时干涉炎症通路及控制炎症的发生发展可能是治疗CD的重要机制。OBJECTIVE This study was to explore the molecular mechanism of Scutellaria Decoction for Crohn′s disease(CD).METHODS The potential active ingredient of Scutellaria Decoction and potential target genes were screened by TCMSP database,and target genes of CD were screened from GEO chip,Genecard,and OMIM database.Cytoscape software was used to construct the network of Scutellaria Decoction-active ingredient-potential target genes,and the key target genes were selected based on topological parameters.GO biological process enrichment and KEGG pathway analysis were performed on the disease and drug intersection targets using the R package clusterprofile.The molecular mechanism of Scutellaria Decoction for CD was analyzed combined with literature.RESULTS There were 65 key targets in the treatment of CD were obtained.Quercetin,kaempferol,naringenin,wogonin may be the main therapeutic CD of Scutellaria Decoction active ingredients,were acted on ESR1,AKT1,IL6,and other targets.They were involved in PI3K-Akt signaling,HIF-1 signaling,IL-17 signaling,TNF signaling and other pathways to play the role of treating CD.CONCLUSION Scutellaria Decoction in treating CD with multiple components,multiple targets and multiple pathways was predicted,meanwhile regulating inflammatory pathways and intervening the occurrence and development of inflammation may be the important mechanism of Scutellaria Decoction in the treatment of CD.
