摘 要:目的制作小型猪自体原位肝移植肝动脉热缺血再灌注动物模型。方法采用巴马小型猪模拟自体原位肝移植手术过程,对照组仅模拟手术过程,观察组在灌注结束后分为肝动脉和门静脉同时开放组、肝动脉缺血1 h组、肝动脉缺血2 h组,观察各组肝功能指标及肝组织结构变化。结果本研究成功制作巴马小型猪自体原位肝移植肝动脉热缺血再灌注模型;随着肝动脉缺血时间的延长,肝细胞缺血缺氧加重,进而导致胆汁淤积加重,并通过肝功能指标及电镜下观察结果得到验证。结论巴马小型猪自体原位肝移植肝动脉热缺血再灌注模型能很好的模拟手术过程,并精确控制肝动脉缺血时间。Objective To establish the model of hepatic artery warm ischemia reperfusion model in the miniature pigs autologous orthotopic liver transplantation.Methods Bama miniature pigs were used to simulate the operation process of autologous orthotopic liver transplantation,while the control group only simulated the operation process.The observation group was divided into simultaneous opening of hepatic artery and portal vein group,hepatic artery ischemia for 1 h group and hepatic artery ischemia for 2 h group after the end of perfusion.The liver function indexes and the change of liver tissue structure of each group were observed.Results In this study,the hepatic artery warm ischemia reperfusion model in the Bama miniature pigs autologous orthotopic liver transplantation was successfully established;with the prolongation of hepatic artery ischemia time,hepatocyte ischemia and hypoxia worsen,this in turn leads to worsening cholestasis,which was verified by liver function indexes and observation results under electron microscope.Conclusion The hepatic artery warm ischemia reperfusion model in the Bama miniature pigs autologous orthotopic liver transplantation can simulate the operation process well and control the time of hepatic artery ischemia accurately.
