别嘌醇对糖尿病并慢性间歇性缺氧大鼠肾损伤的保护作用

摘  要:目的研究别嘌醇(ALLO)对糖尿病并慢性间歇性缺氧(CIH)大鼠肾损伤的保护作用。方法将雄性Wistar大鼠随机分为正常对照组(10只)和糖尿病组(40只),糖尿病组造模成功后分为糖尿病对照组(A组)、CIH模型对照组(B组)、小剂量组[ALLO 20 mg/(kg·d),C组]、大剂量组[ALLO 40 mg/(kg·d),D组]行CIH模型造模,每组10只。检测并分析各组的尿微量白蛋白(mALB)、胱抑素C(CysC)、尿酸(UA)、糖化血红蛋白(HbA1c)、血管内皮生长因子(VEGF)、超氧化物歧化酶(SOD)、丙二醛(MDA)、黄嘌呤氧化酶(XO)蛋白表达水平。结果B组的mALB、CysC、UA、HbA1c、VEGF及肾组织MDA、XO表达水平明显高于正常对照组及A、C、D组,SOD水平明显低于正常对照组及A、C、D组(P<0.05)。D组血mALB、CysC、HbA1c水平显著低于C组,SOD水平显著高于C组,差异具有统计学意义(P<0.05)。结论糖尿病并CIH大鼠的肾损伤与氧化应激及VEGF有关。ALLO对糖尿病并CIH大鼠的血VEGF和氧化应激反应有抑制作用,且随剂量的增大,其对氧化应激反应的抑制加强,进而能够减轻糖尿病并CIH大鼠的肾损伤。Objective To investigate the protective effect of allopurinol(ALLO)on renal injury in rats with diabetes and chronic intermittent hypoxia(CIH).Methods A total of 50 male Wistar rats were randomly divided into normal control group(10 rats)and diabetes group(40 rats).After successful modeling,the diabetic group were divided into diabetes control group(group A),CIH model control group(group B),low-dose group[ALLO 20 mg/(kg·d),group C],and high-dose group[ALLO 40 mg/(kg·d),group D]to receive CIH model,with 10 rats in each group.The expression levels of urinary microalbumin(mALB),Cystatin C(CysC),uric acid(UA),glycosylated hemoglobin(HbA1c),vascular endothelial growth factor(VEGF),superoxide dismutase(SOD),malondialdehyde(MDA)and xanthine oxidase(XO)protein were detected and analyzed.Results The levels of mALB,CysC,UA,HbA1c,VEGF,MDA and XO in the renal tissues of the group B were significantly higher than those of the normal control group and the group A,C and D,while the SOD level was significantly lower than that of the normal control group and the group A,C and D,the differences were statistally significant(P<0.05).The levels of mALB,CysC and HbA1c in the group D were significantly lower than those in the group C,and the level of SOD was significantly higher than that in the group C,the differences were statistally significant(P<0.05).Conclusion The renal injury in rats with diabetes and CIH is related to oxidative stress and VEGF.ALLO has inhibitory effect on blood VEGF and oxidative stress response in rats with diabetes and CIH,and its inhibition on oxidative stress response is enhanced with the increase of dose,so as to reduce kidney injury in rats with diabetes and CIH.
