摘 要:目的探讨4种不同贮存条件对血清白介素8(interleukin-8,IL-8)测定结果的影响。方法随机采集20位受试者的血液标本,每位血液标本为4份,分别为室温不分离血清、室温分离血清、4℃不分离血清、4℃分离血清,再分别于0.5、1、3、5、7、9、11、13、18 h和24 h测定IL-8水平。结果与室温不分离血清组0.5 h比较,5 h内各组检测结果无明显变化(P>0.05),7 h以上各组差异有统计学意义(P<0.05),随着时间的延长差异越显著,且上升幅度无规律;室温分离血清、4℃不分离血清、4℃分离血清条件下,IL-8测得的水平无明显变化(P>0.05)。结论室温不分离血清组血液在5 h之内IL-8检测结果差异无统计学意义,7 h后随时间延长逐渐增高,差异有统计学意义,而在室温分离血清、4℃不分离血清、4℃分离血清条件下标本在24 h内检测不会受时间影响。不同标本保存条件对IL-8实验室检测结果的准确性和可靠性影响较大。如需延时检测,建议分离血清或低温保存,才能够辅助临床进行准确的诊断和有效的治疗。Objective The stability of samples is the key to obtain reliable results in laboratory testing.The goal of the current study is to explore the effect of four different storage conditions on the determination of interleukin-8(IL-8)in serum.Methods Eighty blood samples from 20 subjects were randomly collected.They were distributed to four groups,which were stored at the conditions as following:unseparated serum at room temperature,unseparated serum at 4℃,separated serum at room temperature and separated serum at 4℃.The samples were detected at 0.5 h,1 h,3 h,5 h,7 h,9 h,11 h,13 h,18 h and 24 h,respectively.Results Compared with the room temperature and 0.5h group,there was no significant change in each group within 5 h,while there was significant difference in each group more than 7 h.Moreover,the difference was getting more significant with the extension of time,and the rising range was irregular,while there was no significant change in the concentration of IL-8 at 4℃,room temperature and 4℃.Conclusion When the samples are placed at room temperature,the test results will gradually increase over time,which will significantly affect the accuracy of the results.However,under the conditions of unseparated serum at 4℃,separated serum at room temperature and separated serum at 4℃,the concentration will not change significantly in 24 h.So arranging a reasonable test time for these samples is necessary to improve the accuracy and reliability of IL-8,which can assist doctors to have accurate diagnose and effective treatment.
