摘  要:目的探讨0.22太赫兹(terahertz,THz)电磁辐射暴露致小鼠神经母细胞瘤细胞株——Neuro-2a细胞损伤的非热效应特点。方法以Neuro-2a细胞作为体外实验暴露模型,采用频率为0.22 THz,平均功率密度为0(Sham组)、25(25 mW/cm^2辐照组)、50 mW/cm^2(50 mW/cm^2辐照组)辐照细胞,辐照时间为5 min,监测辐照引起的细胞培养基温度变化;于暴露后24 h检测细胞增殖(EdU阳性细胞比例和细胞活力)、凋亡(γH2AX、TUNEL阳性细胞比例、Bax与Bcl2基因表达水平);于暴露后48 h分析细胞突触生长(突触长度、分支数目及Tubb3与Syp基因表达水平)的变化情况。结果与Sham组比较,两辐照组细胞培养基温度升高在0.1~0.4℃;EdU阳性细胞比例、细胞活力、γH2AX和TUNEL阳性细胞比例及Bax、Bcl2基因表达水平均无明显改变;仅50 mW/cm^2辐照组细胞突触长度和分支数目较Sham组明显减少(P<0.01),Tubb3和Syp基因表达也明显降低(P<0.05)。结论THz辐射暴露以非热效应为主,可抑制Neuro-2a细胞突触生长,而不影响细胞增殖和凋亡,提示神经细胞突触生长是THz辐射产生神经生物效应的敏感靶标。Objective To explore the features of non-thermal effects of cell injury induced by terahertz(THz)radiation at a frequency of 0.22 THz,in a mouse neuroblastoma cell line,Neuro-2 a.Methods The cultured Neuro-2 a cells were exposed to 0.22 THz irradiation at average power densities of 0(sham group),and 25 and 50 mW/cm^2(radiation groups),for 5 min,respectively.The temperature changes of culture medium were monitored.The cell proliferation and apoptosis and the mRNA expression of Bax and Bcl2 at 24 h after THz exposure were detected by measuring the ratio of EdU^+cells,CCK-8 assay for cell viability,immunofluorescence assay forγH2 AX^+cells,TUNEL assay,and real-time PCR.The neurite length,number of branch points,and mRNA expression of Tubb3 and Syp were analyzed in the Neuro-2 a cells at 48 h after THz exposure.Results Compared with the sham group,the temperature of culture medium in the radiation groups was increased by a range from 0.1 to 0.4℃.There were no significant differences in the cell viability,ratios of positive cells for EdU,γH2 AX and TUNEL,or the mRNA expression levels of Bax and Bcl2.The neurite length and number of branch points were obviously reduced(P<0.01),and the mRNA levels of Tubb3 and Syp were also notably down-regulated(P<0.05)in the 50 mW/cm^2 group.Conclusion THz radiation mainly exhibits non-thermal effects,and can inhibit neurite outgrowth but not affect the proliferation and apoptosis in Neuro-2 a cells.Our findings suggest that neurite outgrowth is a sensitive target of neurobiological effects after THz radiation exposure.
