养心定悸胶囊预处理对大鼠心肌缺血再灌注损伤的保护作用

摘　　要：目的探讨养心定悸胶囊预处理对大鼠心肌缺血再灌注损伤的保护作用。方法将36只健康雄性SD大鼠随机分为3组:假手术组仅穿线不结扎;模型组制备心肌缺血再灌注模型;养心定悸组再灌注前给予养心定悸溶液灌胃。所有大鼠灌注结束后,使用氯化三苯基四氮唑(TTC)染色计算大鼠心肌梗死面积,检测心肌坏死标志物肌酸激酶同工酶(CK-MB)和乳酸脱氢酶(LDH)含量。酶联免疫吸附试验(ELISA)测定血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、超氧化物歧化酶(SOD)和丙二醛(MDA)水平。结果与假手术组相比,模型组和养心定悸组大鼠心肌梗死面积明显增加(P<0.05),养心定悸组心肌梗死面积较模型组明显减小(P<0.05)。与假手术组比较,模型组与养心定悸组血清CK-MB、LDH水平均明显增高(P<0.05);养心定悸组血清CK-MB、LDH水平较模型组明显降低(P<0.05)。模型组与养心定悸组TNF-α、IL-6、MDA水平较假手术组明显升高(P<0.05),养心定悸组TNF-α、IL-6、MDA水平明显低于模型组(P<0.05)。模型组与养心定悸组SOD水平较假手术组明显降低(P<0.05),养心定悸组SOD水平高于模型组(P<0.05)。结论养心定悸胶囊预处理对大鼠心肌缺血再灌注损伤具有预防作用,可能与其抗炎、抗氧化作用有关。Objective To investigate the effect of pretreatment with Yangxin Dingji capsule on myocardial ischemia reperfusion injury in rats.Methods Male Sprague-Dawley(SD)rats were randomly divided into three groups:sham group,model group,and Yangxin Dingji capsule(YXDJ)group.The left anterior descending(LAD)was only exposed but not ligated in sham group.Except for sham group,the rat models of myocardial ischemia reperfusion injury were established in all other groups,and the rats in YXDJ group were administered Yangxin Dingji capsule before reperfusion.Myocardial infarct size was measured by TTC method.The levels of creatine kinase-MB(CK-MB)and lactic dehydrogenase(LDH)were measured.The levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),superoxide dismutase(SOD),and malondialdehyde(MDA)were measured by ELISA.Results Compared with sham group,the myocardial infarct sizes in model group and YXDJ group were significantly increased(P<0.05),and the infarct size in YXDJ group was lower than that in the model group(P<0.05).The levels of CK-MB and LDH in I/R group and YXDJ group were significantly higher than those in sham group(P<0.05),and the levels of CK-MB and LDH in YXDJ group was lower than those in model group(P<0.05).The levels of TNF-α,IL-6,and MDA in model group and YXDJ group were significantly increased compared with that in sham group(P<0.05),and the levels of TNF-α,IL-6,and MDA in the YXDJ group were significantly lower than those in model group(P<0.05).The levels of SOD in model group and YXDJ group were significantly lower than that in sham group,and the level of SOD in YXDJ group was higher than that in model group(P<0.05).Conclusion The pretreatment of Yangxin Dingji capsule could prevent the myocardial ischemia reperfusion injury in rats,which may be related to its anti-inflammatory and anti-oxidation functions.
