摘  要:目的建立双波长HPLC法探究柴胡和白芍醋炙前后组方对经方四逆散中5种成分含有量的影响。方法样品70%甲醇溶液的分析采用Kromasil C(18)色谱柱(4.4 mm×250 mm,5μm);流动相乙腈-水,梯度洗脱;体积流量1 mL/min;柱温40℃;检测波长210 nm(柴胡皂苷D)和240 nm(柴胡皂苷B2、芍药苷、甘草苷、橙皮素)。结果四逆散及醋炙品组方四逆散中的柴胡皂苷D、柴胡皂苷B2、芍药苷、甘草苷、橙皮素分别在6.83~1 334、2.12~828、1.044~1 088、1.158~1 206、1.079~1 124μg/mL范围内线性关系良好。四逆散中柴胡皂苷D、芍药苷、甘草苷、橙皮素的平均加样回收率99.85%~101.50%,RSD 0.68%~2.79%;醋炙品组方四逆散中柴胡皂苷D、柴胡皂苷B2、芍药苷、甘草苷、橙皮素的平均加样回收率100.60%~102.76%,RSD均小于0.74%~3.34%。醋炙品组方四逆散中柴胡皂苷B2的含有量显著升高,柴胡皂苷D、甘草苷、橙皮素的含有量变化与四逆散中的柴胡皂苷D、甘草苷、橙皮素相比具有统计学差异,四逆散及醋炙品组方四逆散中芍药苷的含有量变化无统计学差异。结论该方法简便准确,专属性强,可初步揭示柴胡和白芍醋炙前后组方四逆散增效的物质基础。AIM To establish a dual wavelength HPLC method for the simultaneous determination of five active constituents in classical formula Sinisan containing Bupleuri Radix and Paeoniae Radix Alba with or without vinegar-processing.METHODS The analysis of methanol extracts of Sinisan were performed on a Kromasil C18 column(4.4 mm×250 mm,5μm).The mobile phase was composed of acetonitrile and water for gradient elution.The detection wavelengths were set at 210 nm(for detection of saikosaponin D)and 240 nm(for detection of saikosaponin B2,paeoniflorin,liquiritin,hesperitin).The flow rate was 1 mL/min and the column temperature was set at 40℃.RESULTS There were good linear relationships among saikosaponin D,saikosaponin B2,paeoniflorin,liquiritin,and hesperitin in Sinisan with or without vinegar-processed Bupleuri Radix and Paeoniae Radix Alba within ranges of 6.83-1334,2.12-828,1.044-1088,1.158-1206,and 1.079-1124μg/mL,respectively.The average recoveries of saikosaponin D,paeoniflorin,liquiritin,and hesperitin in Sinisan were 99.85%-101.50%,respectively,and the RSDS of each component were 0.68%-2.79%.The average recoveries of saikosaponin D,saikosaponin B2,paeoniflorin,liquiritin,and hesperitin in Sinisan containing vinegar-processed Bupleuri Radix and Paeoniae Radix Alba were 100.60%-102.76%,respectively,and the RSDS of each component were 0.74%-3.34%.Sinisan containing vinegar-processed Bupleuri Radix and Paeoniae Radix Alba shared significantly increased content of saikosaponin B2,and significant content differences in saikosaponin D,liquiritin,and hesperitin except for paeoniflorin in contrast to Sinisan.CONCLUSION With strong specificity,this simple and accurate method can preliminarily reveal the material basis of the synergistic effects of Sinisan due to vinegar-processed Bupleuri Radix and Paeoniae Radix Alba.
