摘　　要：目的:分析GATA-3、胸腺基质淋巴细胞生成素(TSLP)和白细胞介素4、5(IL-4、IL-5)在嗜酸粒细胞性慢性鼻窦炎伴有鼻息肉(ECRSwNP)和非嗜酸粒细胞性慢性鼻窦炎伴有鼻息肉(non-ECRSwNP)患者中的表达及意义。方法:应用免疫组织化学和实时荧光定量PCR试验分别在蛋白和mRNA水平检测GATA-3和TSLP在25例ECRSwNP,23例non-ECRSwNP和20例正常下鼻甲组织中表达,同时采用酶联免疫吸附试验(ELISA)检测IL-4和IL-5的表达。结果:GATA-3 mRNA和蛋白表达在ECRSwNP组均高于non-ECRSwNP组和对照组(P<0.05),TSLP mRNA和蛋白表达在ECRSwNP组表达高于non-ECRSwNP组和对照组(P<0.05)。IL-4和IL-5含量在ECRSwNP组均高于non-ECRSwNP组和对照组(P<0.05)。GATA-3与IL-4和IL-5呈正相关(P<0.05),TSLP与IL-4和IL-5呈正相关(P<0.05),GATA-3与TSLP呈正相关(P<0.05)。结论:TSLP促进GATA-3表达和IL-4和IL-5的产生,GATA-3表达诱导IL-4和IL-5表达增高。GATA-3和TSLP参与了嗜酸粒细胞性CRSwNP的发生发展。Objective:To evaluate expression and clinical implications of GATA-3、TSLP、IL-4 and IL-5 in patients with eosinophilic and non-eosinophilic chronic rhinosinusitis with nasal polyps.Methods:Immunohistochemistry and quantitative RT-PCR were used to detect the mRNA and protein expression of GATA-3 in 25 patients with ECRSwNP,23 patients with non-ECRSwNP and 20 control with normal inferior turbinate tissues.IL-4 and IL-5 were detected by enzymy-linked immunosorbent assay(ELISA).Results:The mRNA and protein expression level of GATA-3 were significantly higher in the ECRSwNP than the non-ECRSwNP group and the control group(P<0.05),the mRNA and protein expression level of TSLP were remarkably higher in the ECRSwNP than the non-ECRSwNP group and the control group(P<0.05);The level of IL-4 and IL-5 were markedly higher in the ECRSwNP than the non-ECRSwNP group and the control group(P<0.05);The expression of GATA-3 was closely correlated with the level of IL-4 and IL-5(P<0.05),TSLP expression was closely correlated with the level of IL-4 and IL-5(P<0.05),GATA-3 expression was closely correlated with the expression of TSLP(P<0.05).Conclusion:TSLP enhances the expression of GATA-3 and production of IL-4 and IL-5,GATA-3 promotes production of IL-4 and IL-5,The GATA-3 and TSLP play an important role in the pathogenesis of ECRSwNP.
