摘 要:目的采用全自动酶免分析仪和酶标板快速孵育器相结合,优化酶联免疫吸附操作流程,缩短检测时间,提高工作效率。方法收集化学发光法检测结果为高(≥250 IU/ml)、中(101~250 IU/ml)、低(0.05~100 IU/ml)值的HBsAg阳性标本及阴性对照各50例,分别用全自动酶免分析仪,以及全自动酶免仪联合酶标板快速孵育器设置孵育时间为常规时间的1/2,1/3,1/4进行检测,比较与化学发光检测结果的差异性。结果与化学发光检测相比较,全自动酶免分析仪对HBsAg的检测结果差异无统计学意义(χ2=0.053,P>0.05),全自动酶免仪联合酶标板快速孵育器分别缩短孵育时间至常规1/2,1/3和1/4对HBsAg检测结果差异均无统计学意义(χ2=0~0.323,均P>0.05)。各方法比较差异均无统计学意义,但其中将快速孵育时间缩短至试剂盒规定时间的1/2时的总样本符合率和敏感度最高。结论全自动酶免分析仪联合酶标板快速孵育器缩短孵育时间至常规的1/2时,显著提高检测结果准确度,同时提高工作效率,在临床标本检测中具有较高的推广应用价值。Objective To explore the method of optimizing the operation flow of enzyme-linked immunosorbent assay(ELISA),shortening incubation time and improving work efficiency by using the combination of rapid incubation method and automatic enzyme immunoassay analyzer.Methods Collected 50 HBsAg negative specimens and positive specimens with high(≥250 IU/ml),medium(101~250 IU/ml),and low(0.05~100 IU/ml)values by chemiluminescence method,respectively.Detection with automatic enzyme immunoassay analyzer and combined detection with automatic enzyme immunoassay analyzer and rapid incubator combined were carried out simultaneously.At the same time,the rapid incubation time was shortened to 1/2,1/3 and 1/4 of the kit’s prescribed time.And then compared the results with the chemiluminescence method.Results Compared with chemiluminescence detection,the results of HBsAg detection by the automatic enzyme free analyzer were not statistically significant.The results of HBsAg detection by the automatic enzyme free analyzer combined with the rapid incubator of enzyme label plate reduced the incubation time to 1/2,1/3 and 1/4,respectively,the difference were not statistically significant.There were no statistically significant differences among the methods,but the total sample coincidence rate and sensitivity were the highest when the rapid incubation time was reduced to 1/2 of the time specified by the kit.Conclusion When the automatic enzyme-free analyzer combined with the rapid incubator of enzyme-label plate shorted the incubation time to 1/2 of the routine,the detection accuracy and work efficiency were significantly improved,which had a high value of popularization and application in clinical specimen detection.
