摘  要:目的探究地黄甙D对四氯化碳诱导的肝纤维化大鼠的保护作用。方法40只SD大鼠随机分成4组:对照组、模型组、地黄甙D20 mg/kg组和地黄甙D60 mg/kg组,每组10只。采用腹腔注射四氯化碳法构建肝纤维化大鼠模型。采用酶联免疫吸附法检测血天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、透明质酸(HA)、层粘连蛋白(LN)、Ⅲ型前胶原(PCⅢ)、Ⅳ型胶原(CⅣ)水平;蛋白质免疫印迹法检测肝组织中天冬氨酸特异性半胱氨酸蛋白酶3(Caspase-3)水平结果与模型组比较,地黄甙D组大鼠AST、ALT、HA、LN、PCⅢ、CⅣ及Caspase-3水平均降低,差异均有统计学意义(P<0.05)。地黄甙D60 mg/k g组大鼠AST、ALT、H A、LN、PCⅢ、CⅣ及Caspase-3水平均低于地黄甙D20 mg/kg组,差异均有统计学意义(P<0.05)。结论地黄甙D对四氯化碳诱导的肝纤维化大鼠有一定保护作用,其作用可能与调控Caspase-3表达有关。Objective To explore the protective effect of rehmainonoside D on carbon tetrachloride-induced liver fibrosis in rats.Methods Forty SD rats were randomly divided into four groups:control group,model group,20 mg/kg rehmainonoside D group and 60 mg/kg rehmainonoside D group,with 10 rats in each group.A rat model of liver fibrosis was established by intraper-itoneal injection of carbon tetrachloride.Blood aspartate aminotransferase(AST),alanine aminotransferase(ALT),hyaluronic acid(HA),laminin(LN),type Ⅲ procollagen(PC Ⅲ),type Ⅳ collagen(C Ⅳ)levels were detected by enzyme-linked immunosorbent assay.Western blotting was used to detect Caspase-3 levels in liver tissues.Results The levels of AST,ALT,HA,LIN,PC Ⅲ,C Ⅳ and Caspase-3 were significantly decreased in rats in the rehmainonoside D groups when compared with those in the model group,and the differences were statistically significant(P<0.05).Those indicators were also much lower in the 60 mg/kg rehmainonoside D group than those in the 20 mg/kg Rehmainonoside D group,with the differences being statistically significant(P<0.05).Conclusion Rehmainonoside D has a protective effect on carbon tetrachloride-induced liver fibrosis in rats,and its effect may be related to the regulation of Caspase-3 expression.
