摘 要:目的探究三种不同离心速度在洗涤红细胞制备中对成品质量的影响。方法选取惠州市中心血站2018年2月至11月期间无偿献血过程收集的悬浮红细胞123袋进行不同速度离心制备洗涤红细胞试验,所选悬浮红细胞ALT>40 U/L,采取随机数表法将123袋悬浮红细胞分为三组,计为中速组、高速组和变速组,每组各41袋。制备洗涤红细胞后检测每组红细胞回收率、血浆蛋白清除率、上清蛋白质含量、白细胞清除率、血红蛋白含量及溶血率情况。结果中速组血红蛋白含量(71.62±6.25)mg/L高于变速组(61.53±3.65)mg/L和高速组(62.49±4.38)mg/L,差异有统计学意义(P<0.05);中速组溶血率(0.43±0.08)%明显低于变速组(0.63±0.09)%和高速组(0.61±0.07)%,差异有统计学意义(P<0.05)。中速组白细胞清除率(92.43±1.69)%明显高于变速组(87.53±1.56)%、高速组(87.36±1.58)%,差异有统计学意义(P<0.05)。结论在符合洗涤红细胞的制备标准前提下,离心速度设为3500r/min制备洗涤红细胞的质量最优。Objective To explore the effect of three different centrifugal speeds on the quality of finished products in the preparation of washed red blood cells.Methods 123 bags of suspended red blood cells collected in Huizhou Provincial Blood Center during the period from February to November 2018 were used to prepare washed red blood cells by centrifugation at different speeds,The ALT of suspended red blood cells was>40 U/L,123 bags of suspended red blood cells were divided into three groups by random number table method,It is divided into medium speed group,high speed group and variable speed group,41 bags in each group.The recovery rate of erythrocyte,plasma protein clearance rate,supernatant protein content,white blood cell clearance rate,hemoglobin content and hemolysis rate of each group were detected after preparation of washed red blood cells.Results The content of hemoglobin in the medium speed group(71.62±6.25)mg/L was significantly higher than that in the variable speed group(61.53±3.65)mg/L and the high speed group(62.49±4.38)mg/L,The difference was significant(P<0.05);The hemolysis rate of the medium-speed group(0.43±0.08)%was significantly lower than that of the variable-speed group(0.63±0.09)%and the high-speed group(0.61±0.07)%,The difference was significant(P<0.05).The clearance rate of white blood cells in the medium speed group(92.43±1.69)%was significantly higher than that in the variable speed group(87.53±1.56)%and the high speed group(87.36±1.58)%,The difference was significant(P<0.05).Conclusion On the premise of meeting the preparation standard of washed red blood cells,The optimum centrifugal speed is 3500 r/min for the preparation of washed red blood cells.
