摘  要:以土壤为材料,经过初筛平板富集培养,分离纯化得到了能正常生长的28株细菌和35株放线菌;再分别进行液体发酵,用改进的硫酸-咔唑法测定胞内、外酶活,筛选出了一株产胞外葡萄糖异构酶活力最高的细菌株菌XJ-27.对该菌株进行形态特征和16S rDNA序列比对分析,并对其胞外葡萄糖异构酶进行了部分性质研究.结果表明:该菌株酶活力为242.1 U·mL^-1,属于芽孢杆菌属(Bacillus),70℃测得最适pH为7.0~7.8;在pH 7.4时其最适反应温度为70~80℃;Mg^2+和Co^2+对其酶活力有显著提高,Mg^2+的效果最明显.After initial screening and on-plate enrichment culture,28 strains of bacteria and 35 strains of actinomycetes were isolated and purified from multiple soil samples.Liquid fermentation was carried out separately,the intracellular and extracellular enzyme activities were determined by improved carbazole-sulfate method.XJ-27,a bacterial strain with the highest activity of extracellular glucose isomerase was selected.XJ-27 was analyzed by morphological characterization and 16S rDNA sequence.Some properties of extracellular glucose isomerase of this strain were also studied.The results indicated that XJ-27 belonged to Bacillus,and its enzyme activity was 242.1 U·mL^-1.The optimum pH was 7.0-7.8,and the optimum reaction temperature was 70-80℃under pH 7.4.Mg^2+and Co^2+could significantly increase the enzyme activity and Mg^2+was the most effective.
