东阿阿胶促进小鼠卵泡发育和卵巢分泌的分子机制

摘　　要：阿胶是一味名贵的滋补中药,对人和动物的生殖具有积极作用。顺铂是一种广泛使用的抗癌药物,具有较强的生殖毒性。为研究阿胶促进动物生殖功能(滋阴功效)的分子机制,同时模拟阿胶对化疗后动物卵巢生殖恢复的效果,试验选取了40只5周龄雌性小鼠进行腹腔注射顺铂,制作动物卵巢损伤模型,同时采用阿胶对卵巢损伤的动物模型进行灌胃处理,进而检测阿胶处理小鼠卵巢细胞抗凋亡状况;试验使用RT-qPCR,蛋白免疫印迹和免疫荧光法检测生殖细胞相关基因的表达。结果表明:阿胶能够促进小鼠卵巢和卵泡发育,降低顺铂对卵巢的毒害作用与炎症反应。顺铂和阿胶共处理组与顺铂处理组相比,小鼠卵巢细胞的抗凋亡作用增强,卵巢中生殖与颗粒细胞凋亡减少,各级卵泡数目增加。综上所述,阿胶能促进卵巢和卵泡的发育(滋阴功效)及生殖细胞相关基因的表达,降低顺铂导致的卵巢损伤,对卵巢细胞凋亡具有较强的抵抗作用。同时,阿胶能降低顺铂诱导的动物卵巢炎症反应。Colla Corii Asini is a precious nourishing traditional Chinese medicine,which has positive effects on human and animal reproduction.Cisplatin is widely used as an anticancer drug with strong reproductive toxicity.In order to study the molecular mechanism of Colla Corii Asini in promoting animal reproductive function(nourishing yin effect)and simulate the effect of Colla Corii Asini on the recovery of animal ovarian reproduction after chemotherapy,40 five-week-old female mice were selected with intraperitoneal injection of cisplatin to make animal ovarian injury models.At the same time,the animal models of ovarian injury were treated with intragastric administration by Colla Corii Asini to test the anti-apoptotic status of ovarian cells in mice.Finally,RT-qPCR,Western Blot and immunofluorescence were used to detect the expression of germ cell-related genes.The results showed that Colla Corii Asini could promote the development of mouse ovaries and follicles,and reduce the toxic effects and inflammation of cisplatin on the ovaries.Compared with the cisplatin treatment group,the anti-apoptotic effect of co-treatment with cisplatin and Colla Corii Asini in mouse ovarian cells was enhanced,the apoptosis of germ cells and granulosa cells in the ovary decreased,and the number of follicles at various levels increased.In summary,Colla Corii Asini can promote the development of ovaries and follicles(nourishing yin effect)and the expression of of germ cell related genes,reduce ovarian damage caused by cisplatin,and have a strong resistance to ovarian cell apoptosis.Simultaneously,Colla Corii Asini can reduce cisplatin-induced ovarian inflammation in animals.
