摘 要:利用随机扩增多态性DNA(RAPD)技术对青岛鳌山卫和红岛的2个四角蛤蜊(Mactra veneriformis)自然群体进行了遗传多样性研究。本研究筛选了15条随机引物,其中有8条引物均存在于2个自然群体中。2个自然群体各36个四角蛤蜊共产生82个扩增片段,即每条引物获得2~6个大小不等的片段。2个群体共检测出37个多态性位点,平均多态位点数5.125个,其中鳌山卫四角蛤蜊多态位比例(0.53)高于红岛的(0.50)。此外,本研究得到6个与四角蛤蜊壳高、壳长、壳宽及体质量呈显著相关的随机扩增多态性DNA(RAPD)位点,有助于利用RAPD标记技术为四角蛤蜊新品种选育。The genetic diversity of two natural populations of Mactra veneriformis in Aoshanwei and Hongdao,Qingdao,were investigated with random amplified polymorphism DNA(RAPD).Herein,15 pairs of random primers were selected,among which 8 pairs of primers were both detected in the 2 natural populations.A total of 82 amplification fragments were generated from all the 36 clams in the two natural population,which means that 2~6 fragments of different sizes were obtained from each pairs of primers.Meanwhile,a total of 37 polymorphic loci were detected in the two populations,with an average of 5.125 loci,and the polymorphic loci ratio(0.53)of M.veneriformis in Aoshanwei was higher than that in Hongdao(0.50).In addition,six RAPD loci with significant correlation with shell height,shell length,shell width and weight of the clams were obtained in this study,which is helpful in providing theoretical basis for the breeding of new varieties of M.veneriformis using the RAPD marker technique.
