摘  要:目的:采用化学模式识别分析,比较研究甘草药材及其炮制品炙甘草炮制前后化学成分的差异。方法:采用HPLC,以乙腈-0.1%甲酸水溶液为流动相进行梯度洗脱,研究甘草药材及其炮制品炙甘草HPLC特征图谱,同时测定8个主要化学成分含量以及醇溶性浸出物、水溶性浸出物。通过t检验、相关性分析、相似度分析、主成分分析(principal component analysis,PCA)以及正交偏最小二乘法-判别分析(orthogonal partial least squares discriminant analysis,OPLS-DA)找寻甘草药材及其炮制品炙甘草的差异性成分。结果:甘草药材及其炮制品炙甘草HPLC特征图谱一致性较好,相似度均>0.90;采用PCA对特征图谱结果进行分析,得分散点图显示甘草药材及炙甘草可被分为2组,表明PCA可区分甘草药材及炙甘草。在PCA结果基础上,以8个主要化学成分含量以及醇溶性浸出物、水溶性浸出物为指标,采用t检验及相关性分析,并结合OPLS-DA,共找到4个差异成分,其中贡献最大的差异成分为醇溶性浸出物和甘草素,炮制后两者含量均显著增加。结论:甘草药材及其炮制品炙甘草化学成分存在变化,醇溶性浸出物及甘草素可作为炙甘草炮制前后质量评价的关键性指标。本研究结果可为炙甘草质量的深入研究及质量标准的制定提供依据。Objective:To study the differences of chemical constituents between Glycyrrhiza uralensis Fisch.and its processed products Glycyrrhizae Radix et Rhizoma Praeparata cum melle by chemical pattern recognition analysis.Methods:The gradient elution of acetonitrile-0.1%formic acid aqueous solution were carried out by HPLC.The characteristic chromatograms of Glycyrrhiza uralensis Fisch.and its processed products Glycyrrhizae Radix et Rhizoma Praeparata cum melle were studied.In addition,the contents of eight main chemical components and the water extractives and ethanol extractives were determined.The t-test,correlation analysis,similarity analysis,principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were employed to reveal the discrimination between different samples.Results:The similarities of HPLC characteristic chromatograms of Glycyrrhiza uralensis Fisch.and its processed products Glycyrrhizae Radix et Rhizoma Praeparata cum melle were above 0.90.The HPLC characteristic chromatograms were analyzed by PCA.According to the PCA score scatter plot,24 batches of samples were classified into 2 groups.The classification result indicated that PCA could achieve the effective differentiation of Glycyrrhiza uralensis Fisch.and its processed products Glycyrrhizae Radix et Rhizoma Praeparata cum melle.On the basis of PCA,the contents of eight main chemical components,water extractives and ethanol extractives were taken as indicators.Four different components were found by t-test and correlation analysis combined with OPLS-DA.Finally,ethanol extractives and liquiritigenin were found as the most important markers,which increased significantly after processing.Conclusion:The chemical constituents of Glycyrrhiza uralensis Fisch.and its processed products Glycyrrhizae Radix et Rhizoma Praeparata cum melle changed obviously.The ethanol extractives and liquiritigenin can be used as key indicators for the quality evaluation of Glycyrrhizae Radix et Rhizoma Praeparata cum melle.The results
