ATRA对糖尿病肾病大鼠模型TGF-β1基因甲基化水平的影响

摘  要:分析全反式维甲酸(all-trans retinoic acid,ATRA)在糖尿病肾病(diabetic nephropathy,DN)大鼠模型中,对转化生长因子-β1(TGF-β1)基因甲基化水平的影响。选取8周龄SD大鼠30只,随机分为对照组、DN组和DN+ATRA组。检测大鼠空腹血糖值、24 h尿蛋白定量、血肌酐及尿素氮水平。观察各组大鼠的肾脏结构及纤维化程度。对大鼠肾脏中TGF-β1基因进行甲基化筛选,检测大鼠肾脏组织中TGF-β1的mRNA水平和蛋白表达。结果显示:ATRA降低了DN大鼠的24 h尿蛋白定量、血肌酐和尿素氮水平;ATRA能减轻DN大鼠肾纤维化程度;ATRA能够提高DN大鼠TGF-β1基因的甲基化水平。研究结果表明:ATRA可能通过影响TGF-β1基因甲基化水平来减轻DN大鼠的肾纤维化程度。We aim to analyze the effects of all-trans retinoic acid(ATRA)on the methylation level of TGF-β1 gene in diabetic nephropathy(DN)rat models.Thirty 8-week-old SD rats are randomly divided into control group,DN group and DN+ATRA group.The fasting blood glucose level,the 24-hour urine protein,serum creatinine and blood urea nitrogen levels are tested to observe the kidney structures and the fibrosis of the rats.The methylation level of TGF-β1 gene in the rats’kidneys is isolated,and the mRNA level of TGF-β1 and the protein expression in kidney tissues are monitored.The results show that ATRA can reduce the 24-hour urine protein,serum creatinine and blood urea nitrogen levels as well as the degree of renal fibrosis in DN rats.Furthermore,ATRA can improve the methylation level of TGF-β1 gene in DN rats.In conclusion,ATRA may reduce the degree of renal fibrosis of DN rats by affecting the methylation level of TGF-β1 gene.
