摘 要:目的:应用网络药理学研究方法挖掘六味地黄丸治疗骨质疏松合并糖尿病的物质基础及其作用机制。方法:通过中药系统药理学技术与分析平台TCMSP、TCMID检索熟地黄、山萸肉、山药、牡丹皮、茯苓、泽泻的主要活性成分,并进行靶点预测。分别通过Drugbank、OMIM、DisGeNET、TTD数据库检索骨质疏松与糖尿病的相关疾病靶点,将药物靶点与两种疾病靶点进行交集,筛选得到共同作用靶点。基于STRING平台构建PPI互作网络模型,并利用Cytoscape软件对该网络进行拓扑学分析得到核心靶点。通过Metascape软件对核心靶点进行GO-BP生物功能富集分析以及KEGG Pathway分析。结果:经数据库分析六味地黄丸中含有活性化合物52个,781个作用靶点,1807个骨质疏松相关疾病靶点、糖尿病1671个,中药与疾病共有靶点110个,筛选出核心靶点37个。GO-BP生物功能富集以及KEGG通路分析发现,六味地黄丸可通过运动的积极调节、细胞因子介导的信号通路、细胞对脂质和脂多糖的反应、对外部刺激的积极调节、调控活性氧代谢过程、调节水解酶活性、调节炎症反应、肌肉细胞增殖、血液循环、血管生成、趋化性调节、生殖发育、葡萄糖稳态等生物学过程以及血管内皮生长因子信号通路、白细胞跨内皮迁移通路、FcεRI信号通路、ErbB信号通路等,发挥其治疗骨质疏松合并糖尿病的作用。结论:本研究从多角度挖掘六味地黄丸治疗骨质疏松合并糖尿病的物质基础及作用机制,为今后开展相关研究提供新的科学依据。Objective:The research methods of network pharmacology are used to explore the material basis and mechanism of the Chinese medicine compound Liuwei Dihuang Wan for treating osteoporosis with diabetes.Methods:Through the Chinese Medicine System Pharmacology Technology and Analysis Platform TCMSP、TCMID database,the main active components of the traditional Chinese medicines of Shoudi(Rehmannia),Shanzhuyu(Dogberry),Shanyao(Chinese Yam),Mudanpi(Peony radices cortex),Fuling(Poria)and Zexie(Alisma)were used,and the target prediction was carried out.The Drugbank,OMIM,DisGeNET and TTD databases were used to retrieve the relevant disease targets of osteoporosis and diabetes.The drug target and the two disease targets are intersected to screen the common target.The STRING platform was used to construct a PPI interaction network model,and the network was topologically analyzed using Cytoscape software to obtain the core target.GO-BP biofunctional enrichment analysis and KEGG pathway analysis were performed on the core targets by Metascape software.Results:The database of Chinese herbal medicine Liuwei Dihuang Wan contains 52 active compounds,781 targets.There are 1807 osteoporosis related disease targets,1671 diabetes targets,and 110 drugs and diseases targets.Through the topological analysis of common targets,37 core targets were selected.GO-BP biological function enrichment and KEGG pathway analysis revealed that Chinese medicine compound Liuwei Dihuang Wan can pass through the positive regulation of locomotion,cytokine-mediated signaling pathway,cellular response to lipid,response to lipopolysaccharide,positive regulation of response to external stimulus,positive regulation of reactive oxygen species metabolic process,positive regulation of hydrolase activity,regulation of inflammatory response,muscle cell proliferation,blood circulation,angiogenesis,chemotaxis,developmental process involved in reproduction,glucose homeostasis and other biological functions and VEGF signaling pathway,Leukocyte transendothelial migratio
