摘 要:利用绿原酸与三氯化铁发生显色反应的原理,建立了一种利用纸基微流控数码成像法快速测定绿原酸含量的方法。利用宽胶带在纸上制作疏水边界,将被测流体限定在预设的亲水区域内,引导流体流入检测区与预加的反应剂进行接触反应,用智能手机记录与待测成分浓度直接相关的颜色,并通过QQ软件读取颜色的红(R)、绿(G)、蓝(B)颜色值,探究了该检测体系的显色条件、绿原酸浓度计算模型等。结果表明:优化后的显色条件依次为2.0μL 4%三氯化铁溶液,2.0μL绿原酸溶液点样体积,室温下反应5 min。在此条件下通过多种数学模型的模拟,发现绿原酸浓度在10~50 mg/mL时与√(ΔR^2+ΔG^2+ΔB^2)呈现较好的线性关系,R^2=0.9972;用该法测定金银花绿原酸含量时,回收率为94.29%~97.92%,相对标准偏差为3.6%。该法快捷、成本低廉、检测要求低,可作为复杂样品中绿原酸含量现场快速测定的补充方法。In this paper,a method for the rapid determination of chlorogenic acid content by paper-based microfluidic digital imaging was established based on the principle of chromogenic reaction between chlorogenic acid and ferric chloride.A wide tape was used to make a hydrophobic boundary on the paper.The measured fluid will be confined in a preset hydrophilic area.Fluid flows were guided into the detection areas to react with the pre-added reactant.And a smart phone was used to record directl color which is directly related to the concentration of the measured component.The red(R),green(G),and blue(B)color values were read through QQ software.And the color rendering conditions of the detection system and the calculation model of chlorogenic acid concentration were explored.The results showed that the optimized color development conditions were 2.0 L 4%ferric chloride solution and 2.0 L chlorogenic acid solution point sample volume in turn,and the reaction was conducted for 5 min at room temperature.On this condition,through a variety of mathematical model simulation,it is found that chlorogenic acid concentration in 10~50 mg/mL with square root has a good linear relationship,R2=0.9972;the recovery of chlorogenic acid of Flos lonicerae was 94.29%~97.92%and the relative standard deviation was 3.6%.This method can be used as a supplement to the field rapid determination of chlorogenic acid content in complex samples.
