益气温阳活血利水方对戊巴比妥钠诱导的H9C2心肌细胞线粒体自噬的影响

摘　　要：目的观察益气温阳活血利水方对戊巴比妥钠诱导H9C2细胞损伤后线粒体自噬的影响。方法戊巴比妥钠诱导建立H9C2心肌细胞自噬模型。实验分为空白对照组、模型组、去乙酰毛花苷组和益气温阳活血利水方低、中、高剂量组,各给药组给予相应药物。运用电子显微镜观察自噬小体、线粒体超微结构,三磷酸腺苷(ATP)试剂盒检测心肌细胞ATP浓度,运用活性氧(ROS)荧光探针测定ROS含量,Western blot检测自噬蛋白Beclin 1的表达。结果与模型组比较,益气温阳活血利水方干预后H9C2细胞线粒体边界较清晰,肿胀缓解,空泡明显减少,ATP含量明显增加(P<0.05),ROS含量减少,自噬小体数量减少;益气温阳活血利水方中、高剂量组H9C2细胞Beclin 1表达明显降低(P<0.05,P<0.01)。结论益气温阳活血利水方减少戊巴比妥钠诱导的H9C2心肌细胞损伤与抑制线粒体自噬密切相关,这可能是其治疗心力衰竭的重要机制之一。Objective To observe the effects of Yiqi Wenyang Huoxue Lishui Prescription on pentobarbital sodium-induced mitophagy in H9C2 cardiomyocytes.Methods An H9C2 cardiomyocyte autophagy model was established by pentobarbital sodium induction method.The experiment was divided into blank control group,model group,deacetylethrin group and Yiqi Wenyang Huoxue Lishui Prescription low-,medium-and high-dosage groups.Each administration group was given relevant medicine.Ultrastructures of autophagosomes and mitochondria were observed using an electron microscope,and ATP concentration of cardiomyocytes was detected by ATP kit.Reactive oxygen species(ROS)fluorescent probes were used to determine ROS content,and Western blot was used to detect the expression of autophagy protein Beclin 1.Results Compared with the model group,the mitochondrial boundary of H9C2 cells was clearer,the swelling of mitochondria was relieved,the vacuoles of mitochondria were significantly reduced,the content of ATP increased(P<0.05),the content of ROS decreased,and the number of autophagosomes decreased after the intervention of Yiqi Wenyang Huoxue Lishui Prescription;the protein expression of Beclin 1 in Yiqi Wenyang Huoxue Lishui Prescription medium-and high-dosage groups decreased significantly(P<0.05,P<0.01).Conclusion Reducing pentobarbital sodium-induced H9C2 cells damage by Yiqi Wenyang Huoxue Lishui Prescription is closely related to the inhibition of mitophagy,which may be one of its important mechanisms for treating heart failure.
