摘  要:目的研究雷公藤红素与吲哚美辛的抗炎作用是否具有协同性及其潜在的作用机制。方法采用脂多糖(LPS)诱导RAW264.7的细胞炎症模型,研究雷公藤红素与吲哚美辛及其联用对细胞炎性介质NO、IL-6、TNF-α和IL-1β的影响,通过Chou-Talalay法评价两者联用是否具有协同抗炎作用,Western blot法研究两者联用对JAK/STAT通路的影响。结果雷公藤红素与吲哚美辛能够协同抑制LPS诱导的NO、IL-6、TNF-α和IL-1β的产生,有效抑制JAK1、p-JAK1、JAK2、p-JAK2、STAT3和p-STAT3的表达。结论雷公藤红素和吲哚美辛联用具有协同抗炎作用,可抑制炎症介质的表达,其作用机制与抑制LPS诱导的RAW264.7细胞产生的炎性因子及JAK1/STAT3和JAK2/STAT3信号通路激活有关,推测其具有治疗类风湿关节炎的潜力。Objective To determine the anti-inflammatory effect of celastrol combination with indomethacin,and their potential mechanism.Methods LPS-induced RAW264.7 cells were used to determine the effect of celastrol,indomethacin and their combination on the production of cellular inflammatory mediators,including NO,IL-6,TNF-α and IL-1β.The synergistic anti-inflammatory effect of the combination was evaluateel by Chou-Talalay,and the effect of the combination on the JAK/STAT pathway by Western blot.Results Celastrol combined with indomethacin synergistically inhibited the production of NO,IL-6,TNF-α and IL-1β induced by LPS,and effectively inhibited the expression of JAK1,p-JAK1,JAK2,p-JAK2,STAT3 and p-STAT3.Conclusion Celastrol combined with indomethacin have a synergistic anti-inflammatory effect.They can inhibit the expression of inflammatory mediators and further suppress the activation of JAK1/STAT3 and JAK2/STAT3 signaling pathway in LPS-induced RAW264.7 cell,which might be used to treat rheumatoid arthritis.
