摘 要:目的观察丹皮酚凝胶剂外用给药对角叉菜胶致大鼠足跖肿胀抗炎作用的影响,并初步探讨其作用机制。方法制备丹皮酚凝胶,并在(25±1)℃条件下储存21 d检测其物理稳定性。48只SD雄性大鼠随机分为空白组,模型组,复方醋酸地塞米松乳膏对照组(醋酸地塞米松15 mg·mg^-1),丹皮酚凝胶高、中、低剂量组(12,6,3 mg·mL^-1)。除空白组和模型组外,其余各组取0.1 g丹皮酚凝胶均匀涂抹于大鼠左侧后足处,并用胶布固定,1次·d^-1,连续给药7 d。末次给药1 h后,除空白组外,其余各组于左后足跖皮下注射1%角叉菜胶溶液0.1 mL,3 h后处死大鼠。排水法测量足跖容积计算足肿胀度;计算脾指数、胸腺指数;检测血清中SOD活性、MDA含量;ELISA法检测足跖皮下组织中IL-1β、IL-6、TNF-α、PGE2的含量。结果与模型组比较,丹皮酚凝胶高、中剂量组能够减轻足肿胀程度(P<0.05 或P<0.01);脾指数和胸腺指数降低(P<0.05);血清中SOD活性显著升高(P<0.01),MDA含量显著降低(P<0.01);足跖皮下组织中IL-1β、TNF-α、PGE2的含量降低(P<0.05或P<0.01);丹皮酚凝胶高剂量组IL-6的含量降低(P<0.05)。结论丹皮酚凝胶可显著改善角叉菜胶诱导的足肿胀程度,其作用机制可能与清除自由基、减轻脂质过氧化损伤,抑制炎症局部组织中炎症因子的合成与释放密切相关。Objective To determine the anti-inflammatory effect of external use of paeonol gel on carrageenan induced plantar swelling in rats,and to explore its mechanism.Methods Totally 48 male SD rats were randomly divided into a blank group,a model group,a compound dexamethasone acetate cream control group,and paeonol gel high,medium and low dose groups (12,6,and 3 mg·mL^-1).Except for the blank group and the model group,0.1 g paeonol gel was evenly applied to the left hind foot of rats in all other groups once daily for 7 days and fixed with tape.One hour after the last administration,except the blank group,all other groups were subtaneously injected 1% carrageenan solution 0.1 mL on the left hind sole.The rats were sacrificed 3 h later.The foot swelling degree was calculated by drainage method,the spleen index and the thymus index were calculated,SOD activity and MDA content in the serum were detected,and IL-1β,IL-6,TNF-α and PGE2 contents in the subcutaneous tissue of foot and sole were detected by ELISA.Results Compared with the model group,in the paeonol gel medium and high dose group the degree of foot swelling was reduced (P<0.05,P<0.01),the spleen index and the thymus index decreased (P<0.05),the SOD activity in the serum increased (P<0.01),MDA content decreased (P<0.01),and the content of IL-1β,TNF-α and PGE2 in the subcutaneous tissue of foot metatarsus decreased (P<0.05,P<0.01),and the content of IL-6 in the high dose of paeonol gel decreased (P<0.05).Conclusion Topical administration of paeonol gel significantly improves the degree of carrageenan induced foot swelling in rats.The mechanism may be related to scavenging free radicals,reducing lipid peroxidation damage and inhibiting the synthesis and release of inflammatory factors in the tissue.
