摘 要:试验旨在探究蜂源抗菌肽Melittin与饲料酸化剂乙酸和柠檬酸的体外协同抑菌效应。试验采用微量肉汤稀释法测定大肠杆菌、肠炎沙门氏菌及金黄色葡萄球菌的最小抑菌浓度(MIC);采用琼脂平板菌落计数法测定最小杀菌浓度(MBC);采用棋盘试验稀释蜂源抗菌肽Melittin与饲料酸化剂测定分级抑菌浓度(FIC)。结果显示:蜂源抗菌肽Melittin、乙酸、柠檬酸分别单独使用对3种常见病原菌均具有抑菌作用,将蜂源抗菌肽Melittin分别与乙酸和柠檬酸联合使用时效果显示为协同效应(FIC index<0.5)和相加作用(0.5≤FIC index≤1)。其中,蜂源抗菌肽Melittin与柠檬酸联合使用对大肠杆菌、肠炎沙门氏菌协同抑菌效应最佳,FIC index为0.1875。蜂源抗菌肽Melittin与乙酸联合使用对大肠杆菌与金黄色葡萄球菌协同效应为相加作用,FIC index为0.5。研究表明,蜂源抗菌肽Melittin与乙酸、柠檬酸联合使用会降低单独使用时对动物病原菌的MIC值,作为饲料添加剂联合应用对抗生素替代具有指导作用。The study was to explore the synergistic antibacterial effect of Melittin,in vitro with acetic acid and citric acid,feed acidizing agent.The minimum inhibitory concentration(MIC)of three common pathogens Escherichia coli,Salmonella enteritis and Staphylococcus aureus were determined by microbroth dilution method.The minimum bactericidal concentration(MBC)was determined by AGAR plate count method.Fractional antimicrobial concentration(FIC)was determined by dilution of Melittin and feed acidizing agent in checkerboard experiment.The results showed that bee derived antimicrobial peptides Melittin,acetic acid and citric acid had antibacterial effects on three common pathogens when used alone,respectively.The synergistic effect(FIC index<0.5)and additive effect(0.5≤FIC index≤1)of bee derived antimicrobial peptides Melittin in combination with acetic acid and citric acid were shown.Among them,when Melittin was used in combination with citric acid,it had the best synergistic antibacterial effect on Escherichia coli and Salmonella enteritis,FIC index was 0.1875.When Melittin was used in combination with acetic acid,the synergistic effect on Escherichia coli and Staphylococcus aureus was additive,FIC index was 0.5,which was also the highest value in the experiment.The results suggested that the combination of bee-derived antimicrobial peptide Melittin with acetic acid and citric acid can reduce the MIC value of animal pathogens when used alone,which has a guiding role in the combination of feed additive and antibiotic substitution.
