摘 要:目的:探讨鼻渊通窍颗粒缓解变应性鼻炎(AR)的作用及相关机制。方法:45只SD大鼠随机分为3组,采取不同干预措施,分别为对照组、AR模型组、鼻渊通窍颗粒治疗组。实验结束后处死大鼠,采用免疫组化法检测大鼠鼻黏膜组织中表面活性蛋白(SP-A)表达情况;ELISA检测各组大鼠血清中相关细胞因子的表达差异;Western blot检测TLR4、NF-κB p65蛋白的表达水平。结果:①对照组、AR模型组、治疗组大鼠鼻黏膜中SP-A平均灰度值分别为80.46±6.74、110.95±8.32、102.47±7.77,差异具有统计学意义(P<0.05);②与对照组相比,模型组大鼠血清IgE、ECP、OVA-sIgE、IL-1β、IL-18、IL-17水平明显降低,IFN-γ、TGF-β1水平明显升高(P<0.05)。而治疗组与对照组的IL-1β、IL-18、IL-17水平差异无统计学意义(P>0.05);③与对照组相比,AR模型组及治疗组大鼠Th1、T bet+Th1细胞亚群比例显著下降(P<0.05),Th2、Th17细胞亚群比例显著升高(P<0.05)。与模型组相比,治疗组大鼠Th1、T bet+Th1细胞亚群比例明显升高(P<0.05),Th2、Th17细胞亚群比例下降(P<0.05);④与对照组相比,模型组大鼠鼻黏膜TLR4、NF-κB p65蛋白表达量均显著升高(P<0.05);与模型组相比,治疗组大鼠鼻黏膜TLR4、NF-κBp65蛋白明显下降(P<0.05),与对照组差异无统计学意义(P>0.05)。结论:鼻渊通窍颗粒可通过提高鼻黏膜SP-A水平调节Th1/Th2/Th17细胞平衡从而对AR发挥治疗作用。Objective:To explore the effect and relative mechanism of BiYuanTongQiao granule in relieving allergic rhinitis(AR).Methods:45 SD rats were randomly divided into 3 groups with different intervention measures:control group,AR model group and BiYuanTongQiao granule treatment group,rats were killed after experiment.Expression of SP-A in nasal mucosa of rats was detected by immunohistochemical;expression of related cytokines in serum of rats in each group was detected by ELISA;expression levels of TLR4 and NF-κB p65 protein were detected by Western blot.Results:①Average gray values of SP-A in nasal mucosa in three groups were 80.46±6.74,110.95±8.32,102.47±7.77,respectively(P<0.05);②Compared with control group,levels of serum IgE,ECP,OVA sIgE,IL-1β,IL-18,IL-17 in model group were significantly lower,and levels of IFN-γ,TGF-β1 were significantly higher(P<0.05).There was no significant difference on levels of IL-1β,IL-18 and IL-17 between treatment group and the control group(P>0.05);③Compared with control group,proportion of Th1,T bet+Th1 cell subpopulation in model group and treatment group were decreased significantly(P<0.05),and proportion of Th2,Th17 cell subpopulation were increased(P<0.05);④Compared with control group,TLR4 and NF-κB p65 protein in nasal mucosa of model group were increased significantly(P<0.05),compared with model group,TLR4 and NF-κB p65 protein in nasal mucosa of treatment group were significantly decreased(P<0.05),while there was no significantly difference with control group(P>0.05).Conclusion:BiYuanTongQiao granule can regulate balance of Th1/Th2/Th17 cells to play a therapeutic role in AR by enhance level of SP-A in nasal mucosa.
