摘 要:目的探讨三七总皂苷对缺氧复氧损伤PC12细胞的保护作用及其机制。方法培养PC12细胞,造成缺氧复氧损伤模型,分为对照组、模型组、三七总皂苷低浓度组及三七总皂苷高浓度组,MTT法观察细胞增殖率,测各组细胞上清液乳酸脱氢酶(LDH)及细胞超氧化物歧化酶(SOD)、丙二醛(MDA)变化,流式细胞技术检测PC12细胞内活性氧簇(ROS)表达,Hoechst33258染色检测凋亡细胞比例,Western blot分析Bcl-2及Bax的变化。结果低、高浓度三七总皂苷处理后,PC12细胞增殖率逐渐提高,与模型组比较差异具有统计学意义(P<0.05)。三七总皂苷低、高浓度组细胞外上清液中LDH水平下降,与模型组比较差异有统计学意义(P<0.05)。三七总皂苷低、高浓度组细胞内SOD水平升高,与模型组比较差异有统计学意义(P<0.05)。三七总皂苷低、高浓度组细胞内MDA水平下降,与模型组比较差异有统计学意义(P<0.05)。与模型组比较,三七总皂苷低、高浓度组细胞凋亡率下降(P<0.05)。三七总皂苷低、高浓度组Bcl-2蛋白表达逐渐升高,与模型组比较差异均有统计学意义(P<0.05);三七总皂苷低、高浓度组Bax蛋白的相对表达量下降,与模型组相比差异有统计学意义(P<0.05)。结论三七总皂苷可以抑制缺氧复氧诱导的PC12细胞氧化损伤及凋亡的发生,在预防和治疗脑组织缺血再灌注损伤疾病方面具有一定的临床意义。Objective To investigate the protective effect and mechanism of panax notoginseng saponins(PNS)on hypoxia/reoxygenation PC12 cells damage.Methods PC12 cells were cultured and hypoxia/reoxygenation models were constructed.They were devided into control group,model group,low concentration PNS group and high concentration PNS group.Cell proliferation rate was detected by MTT colorimetry,the changes of lactate dehydrogenase(LDH)in cell supernatant,superoxide dismutase(SOD)and malondialdehyde(MDA)in cells were measured,reactive oxygen species(ROS)expression inside the cells were detected by flow cytometry technology,apoptotic cells proportion were detected by Hoechst33258 staining,the expression of Bcl-2 and Bax were tested by western blot.Results The proliferation rates of PC12 cells elevated after treatment with low or high concentration of PNS,compared with the model group,the differences were statistically significant(P<0.05).The levels of LDH in the extracellular supernatant of the PNS low and high concentration groups decreased,compared with the model group,the differences were statistically significant(P<0.05).Compared with the model group,the levels of SOD in PNS low and high concentration groups elevated,the differences were statistically significant(P<0.05).Compared with the model group,the levels of MDA in PC12 cells of PNS low and high concentration groups decreased respectively,the differences were statistically significant(P<0.05).Compared with the model group,cells apoptosis rates in PNS low and high concentration groups decreased significantly(P<0.05).The expressions of Bcl-2 protein in PNS low and high concentration groups increased gradually,compared with the model group,the differences were statistically significant(P<0.05);The relative expressions of Bax protein in PNS low and high concentration groups were significantly lower,compared with the model group,the difference was statistically significant(P<0.05).Conclusion PNS can inhibit the hypoxia/reoxygenation-induced oxidative damage and apoptosis
