摘  要:目的探讨加味当归贝母苦参丸对慢性非细菌性前列腺炎(CNP)大鼠前列腺组织炎症因子的影响。方法取雄性SD大鼠60只,分为空白组、模型组、加味当归贝母苦参丸低、中、高剂组、对照组,采用注射前列腺蛋白提纯液和完全弗氏佐剂法造模,各组均灌胃给药或生理盐水,1次/d,灌胃4周。给药结束观察大鼠前列腺组织形态学的变化,ELISA法检测前列腺组织IL-1β、IL-10、TNF-a水平。结果模型组前列腺组织炎症明显,加味当归贝母苦参丸组及前列倍喜胶囊组的病理损伤均有不同程度的改善。模型组及低剂量组前列腺组织IL-1β、TNF-a水平明显增高(P<0.01);加味当归贝母苦参丸中、高剂量及前列腺倍喜胶囊组IL-1β、TNF-a水平明显低于模型组(P<0.01)。模型组前列腺组织IL-10水平高于空白组(P<0.01);加味当归贝母苦参丸高剂量组及前列倍喜胶囊组IL-10水平均高于模型组(P<0.01)。结论实验性CNP大鼠模型前列腺组织炎症明显,IL-1β、TNF-a水平升高,IL-10轻度升高;加味当归贝母苦参丸能降低前列腺组织IL-1β、TNF-a水平,提高IL-10水平,改善前列腺组织的病理损伤,这可能是加味当归贝母苦参丸改善前列腺炎患者症状的分子机制之一。Objective To explore the effect of Danggui Beimu Kushen pill on inflammatory factors in prostatic tissue of rats with chronic nonbacterial prostatitis(CNP).Methods 60 male SD rats were randomly selected as control group,model group,Danggui Beimu Kushen pill high,medium and low dose group and Qianlie Beixi capsule group.Rat chronic prostatitis model takes the ventral prostate lobe to produce injection of complete freund's adjuvant method,after the model is successfully established,total groups were given the same dose of saline.Rats in the above groups were given drugs by gavage for 1 time/day,with continuous drug intervention for 4 weeks.After the administration,the morphological changes of the prostate tissue of each group were observed under the light microscope.Results Inflammation of prostate tissue was obvious in the model group,the pathological damage of Danggui Beimu Kushen pill group and Qianlie Beixi capsule was improved to different degrees.IL-1βand TNF-αwere significantly increased in model group and low-dose group.Compared with control group,the difference was statistically significant(P<0.01).The levels of IL-1βand TNF-αin the Dangguibeimukshen decocetion high,medium group and Qianlie Beixi capsule group were significantly lower than those in the model group(P<0.01).Compared with control group,the level of IL-10 in prostate tissue of the model group was significantly higher than that of the control group(P<0.01).The level of IL-10 in both high dosage groups of Dangguibeimukshen decocetion and Qianliebeixi capsule group was higher than that in the model group(P<0.01).Conclusion the inflammation in prostate tissue of experimental CNP rat model was obvious,the levels of IL-1βlevel and TNF-αwere increased,IL-10 was increased slightly,Dangguibeimukshen decocetion can reduce the level of IL-1βin prostate tissue and TNF-α,increase the level of IL-10,and improve the pathological injury of prostate tissue.
