摘 要:为优选大孔树脂对密花香薷全草中总黄酮分离纯化的最佳工艺,以密花香薷全草中总黄酮吸附率、解析率为指标,采用静态和动态吸附-解析实验,结合响应面法,发现DM130型大孔树脂在以下工艺条件下对密花香薷全草中总黄酮的纯化效果最佳:上样液总黄酮为3.50 g·L^-1,上样体积为3 BV,上样液pH为4,流速为1 mL·min-1。洗脱剂为70%乙醇,洗脱剂用量为4 BV,流速为1 mL·min-1。总黄酮的吸附率为89.20%,解析率为89.66%,回收率为77.85%。且纯度由40.96%提升到87.94%。该方法稳定可靠,可用于大孔树脂分离纯化密花香薷全草中的总黄酮。To optimize the best process for the separation and purification of total flavonoids from the whole herb of Elsholtzia densa Benth.by macroporous resin,in this work,we make use of the adsorption rate and resolution factor as the evaluation criteria in static and dynamic adsorption analytical experiments,combined with response surface methodology.We found that the DM130 macroporous resin has the best purification performance on total flavonoids from the whole herb of Elsholtzia densa Benth.The process conditions are:3.50 g·L^-1 total flavonoids concentration of the loading solution,3 BV volume of the loading solution,pH value at 4,flow velocity 1 mL·min-1,eluent 70%ethanol,elution volume 4 BV,and flow velocity 1 mL·min-1.The adsorption rate of the total flavonoids is 89.20%,the resolution factor is 89.66%,and the recovery rate is 77.85%.The degree of purity is increased from 40.96%to 87.94%.This method is stable and reliable,as presented in this work,which can be effectively used to separate and purify the total flavonoids from the whole herb of Elsholtzia densa Benth.by macroporous resin.
