摘  要:为鉴定自然感染的绵羊肺腺瘤肿瘤组织与绵羊健康肺组织之间的差异表达基因(DEGs),本研究采用Illumina Hiseq 4000高通量RNA-seq测序技术对自然感染的绵羊肺腺瘤肿瘤组织及绵羊健康肺组织进行转录组测序和生物信息学分析。参考绵羊基因组(Ovisaries3.1.88)对测序结果进行拼接与质量评估,测序数据处理后获得DEGs,上调的DEGs和下调的DEGs分别利用GO数据库进行富集分析。结果显示:自然感染的绵羊肺腺瘤肿瘤组织与绵羊健康肺组织比较,DEGs共有366个,其中上调的DEGs 154个,下调的DEGs 212个。通过GO功能富集分析,表达上调的DEGs主要富集于L-亮氨酸跨膜转运蛋白活性、铜还原酶活性、NADH脱氢酶活性、NAD+二磷酸酶活性、磷酸蛋白酶活性的负调控、Nem1-Spo7磷酸酶复合物等生物学功能;表达下调的DEGs主要富集于Ⅲ型转化生长因子β受体结合、DNA导向聚合酶活性、胃气化趋同延伸、面部发育等生物学功能。随机选取7个DEGs利用RT-qPCR方法验证,结果显示这7个DEGs的转录水平与上述结果一致,进一步表明转录组分析数据可信。本实验分析并验证了自然感染后期的绵羊肺腺瘤肿瘤组织的DEGs,为研究自然感染的绵羊肺腺瘤相关分子致瘤机制提供了数据支持。To identify the differentially expressed genes(DEGs)in the lung tissue of ovine pulmonary adenocarcinoma in comparsion with sheep healthy lung tissue,these tissues were performed transcriptome sequencing by using Illumina Hiseq 4000 high-throughput RNA-seq sequencing technology,and the bioinformatics analysis was carried out for the resulting sequinces.DEGs were screened by data processing for the sequences after fragment assembly and quality assessment in use of the sheep genome(Ovisaries3.1.88)as the reference sequence.The up-regulated DEGs and down-regulated DEGs were separately enriched using the GO database.As a result,there were 366 DEGs in total including 154 up-regulated DEGs and 212 down-regulated DEGs.A functional enrichment analysis was carried out with bioinformatics,and the up-regulated DEGs are mainly enriched in the negative regulation of L-leucine transmembrane transporter activity,copper reductase activity,NADH dehydrogenase activity,NAD+bisphosphatase activity,phosphoprotease activity,Nem1-Spo7 phosphatase complex and other biological functions;the down-regulated DEGs are mainly enriched in biological functions such as typeⅢtransforming growth factorβreceptor binding,DNA-directed polymerase activity,gastric gasification convergence extension,and facial development.Seven DEGs were selected randomly and used for RT-qPCR assay.the results showed that the transcriptome analysis data was releatively credible.In this study,the DEGs were confirmed in the lung tissue of ovine pulmonary adenocarcinoma during the late stage of natural infection,it will be a good support of data for the futher study on the molecular mechanism of this disease.
