一株牛源金黄色葡萄球菌噬菌体VB-SavM-JYL02生物学特性及基因组学研究

摘　　要：为了解金黄色葡萄球菌(S.aureus)噬菌体生物学及其基因组学特性,本研究利用牛源S.aureus从污水中分离得到一株烈性噬菌体,命名为VB-SavM-JYL02。形态学分析显示该噬菌体属于肌尾病毒科(Myoviridae),基因组全长141384 bp,编码218个开放阅读框,未发现其与细菌毒力、抗生素抗性以及溶原相关的基因。生物学特性研究结果显示,该噬菌体最佳感染复数为0.01,潜伏期20 min,爆发期约为20 min,裂解周期约40 min,表明该噬菌体具有很强的杀菌效率。此外,噬菌体VB-SavM-JYL02具有较宽的宿主谱(64%)。噬菌体全基因组同源性比对分析显示VB-SavM-JYL02与已报道的9个噬菌体存在极其相似的基因模块排布顺序。这些结果表明该噬菌体可能具有用于S.aureus感染治疗的潜力。本研究为噬菌体进一步用于奶牛乳腺炎的治疗研究奠定了基础。To understand the biology characteristics and genomics of Staphylococcus aureus(S.aureus)phages,a bacteriophage of S.aureus was isolated and named VB-SavM-JYL02.Morphological analysis showed that the bacteriophage belonged to myoviridae.The whole genome of bacteriophage VB-SavM-JYL02 is 141384bp in length,encoding 218 open reading frames.No genes related to bacterial virulence,antibiotic resistance and lysogen were found.The results of biological characteristics showed that the optimal infection complex number of the phage was 0.01,latent period was 20 minutes,the outbreak period was about 20 minutes,and the lysis period was about 40 minutes,which indicated that the phage had a high bactericidal efficiency.In addition,the bacteriophage VB-SavM-JYL02 has a broad host spectrum(64%).The homology analysis of whole genome of phage showed that VB-savm-JYL02 existed with 9 reported phages and its similar gene module arrangement sequence.These results indicated that the phage might have a potential for use as a treatment candidate for S.aureus infections.This study laid the foundation for the further use of bacteriophage in the treatment of dairy cow mastitis.
