摘　　要：目的探讨胃肠安丸治疗溃疡性结肠炎的可能作用机制。方法60只C57BL/6小鼠随机选取10只为正常组,其余小鼠给予2.5%葡聚糖硫酸钠(DSS)水溶液自由饮用7天复制UC小鼠模型,将造模成功的50只小鼠再随机分为模型组、美沙拉嗪组和胃肠安丸高、中、低剂量组,每组10只。胃肠安丸低、中、高剂量组分别给予胃肠安丸混悬液0.015、0.03、0.06g/(kg·d)灌胃,美沙拉嗪组给予美沙拉嗪混悬液0.52g/(kg·d)灌胃,正常组和模型组给予0.5ml纯净水灌胃,每日1次,连续10天。每天观察各组小鼠体质量、大便性状及便血情况,并计算疾病活动指数(DAI)。药物干预结束后处死小鼠,测量结肠长度,对结肠组织进行HE染色,观察远端结肠组织病理形态学变化,流式细胞术检测小鼠脾脏调节性T细胞(Treg)、辅助性T细胞17(Th17)百分比及比值。结果干预后与模型组和胃肠安丸低剂量组比较,美沙拉嗪组、胃肠安丸高剂量组小鼠体重差值增加,DAI评分差值升高(P<0.05)。与正常组比较,模型组小鼠结肠长度减少,病理学评分升高,Treg细胞百分比、Treg/Th17值降低,Th17细胞百分比升高(P<0.05);与模型组比较,美沙拉嗪组、胃肠安丸高剂量组小鼠结肠长度增长,病理学评分降低,Treg细胞百分比、Treg/Th17值升高,Th17细胞百分比降低(P<0.05)。美沙拉嗪组、胃肠安丸高剂量组各指标组间比较差异无统计学意义(P>0.05)。结论胃肠安丸在改善DSS诱导的溃疡性结肠炎小鼠腹泻、便血、体质量下降等症状和修复肠道黏膜损伤方面与美沙拉嗪功效相近,其作用机制可能与调节Treg/Th17免疫失衡有关。Objective To explore the possible mechanism of Weichang′an Pill(胃肠安丸)in the treatment of ulcerative colitis.Methods A total of 10 in 60 C57 BL/6 mice were randomly selected as the normal group,and the remaining mice were given 2.5%dextran sodium sulfate(DSS)aqueous solution drinking freely for 7 days to replicate the UC model.Fifty successfully modeled mice were randomly divided into a model group,a mesalazine group and a Weichang′an Pill high,medium and low dose groups,with 10 rats in each group.The low,medium,and high-dose Weichang′an Pill groups were given the Weichang′an Pill suspension 0.015,0.03,0.06 g/(kg·d)respectively by gavage,and the mesalazine group was given Mesalazine suspension 0.52 g/(kg·d)by gavage,the normal group and the model group were given 0.5 ml of purified water once a day for 10 consecutive days.The body weight,stool characteristics and stool blood in each group of mice were observed every day,and the disease activity index(DAI)was calculated.After the drug intervention,the mice were sacrificed,the colon length was measured,the colon tissue was stained with HE,and the pathological changes of the distal colon tissue were observed.Flow cytometry was used to detect the regulatory T cells(Treg)and helper T cells of the mouse spleen 17(Th17)percentage and ratio.Results After intervention,compared with the model group and the Weichang′an Pill low-dose group,the body weight difference of mice in the mesalazine group and the Weichang′an Pill high-dose group increased,and the difference in DAI score increased(P<0.05).Compared with the normal group,the mouse colon length in the model group decreased,the pathological score increased,the percentage of Treg cells,Treg/Th17 values decreased,and the percentage of Th17 cells increased(P<0.05);compared with the model group,the mesalazine group,the colon length of mice in the Weichang′an Pill high-dose group increased,the pathological score decreased,the percentage of Treg cells and Treg/Th17 value increased,and the percentage of T
