当飞利肝宁胶囊对急性肝损伤模型小鼠肝组织锌指蛋白A20表达及NF-κB信号通路的影响

摘　　要：目的探讨当飞利肝宁胶囊对急性肝损伤的保护作用及机制。方法30只C57BL/6小鼠随机分为空白组、模型组和当飞利肝宁组,每组10只。当飞利肝宁组给予当飞利肝宁胶囊0.225g/(kg·d)灌胃10天,空白组和模型组小鼠给予等剂量的生理盐水灌胃。药物干预结束后,模型组和当飞利肝宁组小鼠给予腹腔注射D-半乳糖胺800mg/kg建立急性肝损伤模型,对照组小鼠腹腔注射100μl生理盐水。24h后取材,检测各组小鼠肝组织病理、肝功能[包括丙氨酸氨基转移酶(ALT)和门冬氨酸氨基转移酶(AST)],血清和肝脏组织匀浆中锌指蛋白A20和肿瘤坏死因子α(TNF-α)表达,肝组织锌指蛋白A20、TNF-α、白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、白细胞介素8(IL-8)mRNA表达及核因子κB p65(NF-κB p65)、磷酸化核因子κB p65(p-NF-κB p65)、核因子κB抑制蛋白α(IκBα)、磷酸化核因子κB抑制蛋白α(p-IκBα)蛋白表达。结果肝组织病理结果显示,模型组小鼠的肝组织炎症细胞浸润明显增加,而当飞利肝宁组小鼠肝脏组织炎症细胞浸润情况较模型组明显改善。与空白组比较,模型组小鼠的血清ALT、AST水平升高,血清和肝组织TNF-α表达升高,肝组织TNF-α、IL-1β、IL-6 mRNA表达及p-IκBα和p-NF-κB p65蛋白表达增加(P<0.05或P<0.01);与模型组比较,当飞利肝宁组小鼠血清ALT、AST水平降低,血清和肝组织锌指蛋白A20表达升高、TNF-α表达降低,肝组织锌指蛋白A20 mRNA表达及NF-κB p65蛋白升高,TNF-α、IL-1β、IL-6 mRNA及p-IκBα和p-NF-κB p65蛋白表达降低(P<0.05或P<0.01)。结论当飞利肝宁胶囊能有效改善小鼠急性肝损伤,上调锌指蛋白A20的表达从而调控NF-κB信号通路的激活可能是其潜在作用机制之一。Objective To explore the protective effect and mechanism of Dangfei Liganning Capsules(当飞利肝宁胶囊)on acute liver injury.Method A total of 30 C57 BL/6 mice were randomly divided into a blank group,a model group and a Dangfei Liganning Capsules group,with 10 mice in each group.Dangfei Liganning Capsules group was given Dangfei Liganning Capsules 0.225 g/(kg·d)by gavage for 10 days,and mice in the blank group and model group were given the same amount of normal saline by gavage.After intervention,mice in the model group and Dangfei Liganning Capsules group were given intraperitoneal injection of D-galactosamine 800 mg/kg to establish acute liver injury models,and mice in the control group were intraperitoneally injected with 100μl of normal saline.Samples were taken 24 hours later to detect liver tissue pathology and liver function including alanine aminotransferase(ALT)and aspartate aminotransferase(AST),zinc finger protein A20 and tumor necrosis factorα(TNF-α)expression in serum and liver tissue homogenate,expression of zinc finger protein A20,TNF-α,interleukin 1β(IL-1β),interleukin 6(IL-6),interleukin 8(IL-8)mRNA and expression of nuclear factorκB p65(NF-κB p65)and phosphorylated nuclear factorκB p65(p-NF-κB p65),nuclear factorκB inhibitory proteinα(IκBα),phosphorylated nuclear factorκB inhibitory proteinα(p-IκBα)protein in liver tissue.Results Liver histopathological results showed that the infiltration of inflammatory cells in the liver tissue of mice in the model group was significantly increased,while the infiltration of inflammatory cells in the liver tissue of mice in the Dangfei Liganning Capsules group was significantly improved compared to the model group.Compared with the blank group,the serum ALT and AST levels of the model group increased,TNF-αexpression in the serum and liver tissue increased,expression of TNF-α,IL-1β,IL-6 mRNA and expression of p-IκBαand p-NF-κB p65 protein in the liver tissue increased(P<0.05 or P<0.01).Compared with the model group,in Dang Feili
