摘 要:目的:研究仙茅多糖辅助流感裂解疫苗对小鼠抵抗流感病毒感染的免疫效应。方法:将健康昆明小鼠140只分为空白对照组、模型对照组、仙茅多糖对照组、疫苗对照组以及仙茅多糖不同剂量组。疫苗对照组以及仙茅多糖不同剂量组于第0、14 d采用流感裂解疫苗皮下注射免疫,仙茅多糖不同剂量组在两次疫苗免疫之前分别使用10、20、40 mg/kg的仙茅多糖连续肌肉注射3 d。第21 d检测各组小鼠血清中特异性IgG、IgG1和IgG2a的含量,同时用流感病毒液20μL/只滴鼻感染小鼠,观察小鼠一般情况及21 d内自然死亡情况,并检测肺脏病毒滴度及病理学变化。结果:与空白对照组相比,模型对照组小鼠有显著流感症状,11 d死亡率100%,模型制备成功;与模型对照组相比,疫苗对照组小鼠肺部病毒滴度显著降低(P<0.01),肺组织炎症有所减轻;与疫苗对照组相比,仙茅多糖与疫苗联合免疫可显著提高小鼠血清流感病毒蛋白特异性总IgG及其亚型IgG1、IgG2a的产生水平(P<0.05或P<0.01),显著降低小鼠肺组织中的病毒滴度(P<0.05),提高感染鼠的体重恢复率和生存率。结论:仙茅多糖能够辅助流感裂解疫苗提高小鼠对致死性流感病毒的抗感染能力,有望成为流感疫苗的候选佐剂。Objective:To observe the immune effect of curculigo orchioides polysaccharide(COP)assisted influenza vaccine against influenza virus infection in mice.Mothods:140 healthy Kunming mice were divided into the control group,the model group,COP group,vaccine group and different doses of COP groups.Mice in vaccine group and different doses of COP groups were subcutaneously immunized with influenza vaccine and immunized again on the 14th day after the first immunization.Simultaneously,COP(40,20,10 mg/kg)were intramuscularly injected for 3 days before the two vaccine immunization.On the 21th day,the contents of specific IgG,IgG1 and IgG2a in serum were detected.At the same time,20μl of influenza virus solution was used to infect mice through nasal inhalation.The general situation and natural death situation of mice were observed,the virus titer was detected and the pathological evaluation of lung tissues was performed.Results:Compared with the control group,the mice in the model group had significant influenza symptoms,and the 11-day mortality rate was 100%,which showed that the models were successful.Compared with the model group,the lung virus titer in the vaccine group was significantly reduced(P<0.01),the lung tissue inflammation was reduced.Compared with the vaccine group,the combined immunization of COP and vaccine significantly improved the production levels of the serum influenza virus protein specific total IgG and its subtype IgG1,IgG2a(P<0.05 or P<0.01),it also significantly reduced the virus titer in the lung tissues of mice(P<0.05),and improved the body weight recovery rate and the survival rate of the infected mice.Conclusion:COP can assist influenza vaccine to improve the anti-infective ability of mice against the lethal influenza virus,which may be a potential adjuvant of influenza vaccine.
