高级别浆液性卵巢癌中CTGF表达及临床意义

摘　　要：目的上皮性卵巢癌是妇科肿瘤致死性居首位的恶性肿瘤,急需发现有效的预测指标和治疗靶点。本研究探讨结缔组织生长因子(connective tissue growth factor,CTGF)和上皮间质转化(epithelial-mesenchymal transition,EMT)相关蛋白在高级别浆液性卵巢癌组织中的表达,分析其与临床病理特征、EMT关系。方法收集2015-01-01-2019-10-30本院收治的高级别浆液性卵巢癌组织标本51例及5例正常卵巢组织标本,采用免疫组化链酶菌抗生素蛋白-过氧化物酶法对组织标本进行CTGF、E-cadherin、Vimentin表达检测。结果高级别浆液性卵巢癌细胞中CTGF、E-cadherin和Vimentin阳性表达率分别为100%、100%和56.9%,正常卵巢细胞中均不表达。CTGF表达与E-cadherin表达呈负相关,r=-0.277,P=0.049;CTGF表达与Vimentin表达呈正相关,r=0.392,P=0.004。CTGF高表达与高级别浆液性卵巢癌患者FIGO分期(χ2=15.300,P<0.001)、是否腹膜转移(χ2=12.101,P=0.001)和是否淋巴结转移(χ2=10.137,P=0.001)有关联。结论CTGF在高级别浆液性卵巢癌中表达增高,且与淋巴结转移、腹膜转移及分期相关。CTGF表达与EMT呈正相关,可能参与卵巢癌EMT过程。OBJECTIVE Epithelial ovarian cancer is the most lethal malignant tumor of gynecological tumors.It is urgent to find effective predictive indicators and therapeutic targets.This study explored the expression of connective tissue growth factor(CTGF)and epithelial-mesenchymal transition(EMT)-related proteins in high-grade serous ovarian cancer tissues,and analyzed their clinical pathological characteristics,EMT relationship.METHODS From January 1,2015 to October 30,2019,51 high-grade serous ovarian cancer tissue specimens and 5 normal ovarian tissue specimens from our hospital were collected,and immunohistochemical Streptomyces antibiotic protein-peroxidase(SP)method was used to detect CTGF,E-cadherin and Vimentin expression in tissue samples.RESULTS The positive expression rates of CTGF,E-cadherin and Vimentin in high-grade serous ovarian cancer cells were 100%,100%and 56.9%,respectively,and were not expressed in normal ovarian cells.CTGF expression was negatively correlated with E-cadherin expression,r=-0.277,P=0.049.CTGF expression was positively correlated with Vimentin expression,r=0.392,P=0.004.High CTGF expression was associated with FIGO stage(χ~2=15.300,P<0.001),peritoneal metastasis(χ~2=12.101,P=0.001)and lymph node metastasis(χ~2=10.137,P=0.001)in patients with high-grade serous ovarian cancer.CONCLUSIONS CTGF expression is increased in high-grade serous ovarian cancer,and it is associated with lymph node metastasis,peritoneal metastasis and stage.CTGF expression is positively correlated with EMT and may be involved in the EMT process of ovarian cancer.
