摘  要:目的研究不同剂量硝酸钕对发育期大鼠海马的氧化损伤水平及对发育期大鼠海马中钙离子(Ca^2+)、钙调蛋白(CAM)、脑源性神经营养因子(BDNF)水平的影响,反应其对发育期大鼠学习记忆能力的影响。方法2019年3月至2020年6月选择SPF级SD性成熟大鼠64只,雌雄各半,1∶1同笼交配,至鼠笼底发现阴栓即为妊娠0 d,进行阴性对照、低、中、高剂量组染毒(对应为去离子水、低、中、高硝酸钕浓度为20、200、2000 mg/100 ml)至分娩哺育仔鼠21 d,继续对仔鼠灌胃至断乳后21 d,然后每窝随机选取雌雄仔鼠各1只,每剂量组16只仔鼠,雌雄各半。采用酶联免疫吸附法(ELISA)测定仔鼠海马组织中超氧化物歧化酶(Superoxide dismutase,SOD),丙二醛(Malondialdehyde,MDA),过氧化氢酶(Catalase,CAT),谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px),抗坏血酸过氧化物酶(Ascorbate peroxidase,APX)以及总抗氧化能力(Total antioxidant capacity,TAOC),Ca^2+、钙调蛋白(calmodulin,CaM)、脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)含量。结果脑的相对重量不同剂量组间比较差异无统计学意义(P>0.05),雌性仔鼠不同浓度组间海马中CAT、MDA、GSH-Px、SOD的酶活性差异有统计学意义(P<0.05),雄性仔鼠不同浓度组间海马中CAT、APX、GSH-Px、MDA、SOD组间比较差异有统计学意义(P<0.05),雌性发育期大鼠海马中CAM、BDNF各剂量组间差异有统计学意义(P<0.05),雄性发育期大鼠海马中CAM各剂量组间差异有统计学意义(P<0.05)。结论长期接触高剂量的硝酸钕会使发育期雌、雄性大鼠海马中CAT、GSH-PX和SOD的酶活性降低,MDA增高。Objective To study the effect of different doses of neodymium nitrate on the level of oxidative damage in the hippocampus of developing rats and the effects of calcium ion(Ca^2+),calmodulin(CAM)and brain-derived neurotrophic factor(BDNF)levels in the hippocampus of developing rats,and its effect on the learning and memory ability of rats during the developmental period.Methods A total of 64 sexually mature SPF SD rats(half males and half females in number)were selected and mated with the cage at a ratio of 1∶1 until the vaginal plug was found at the bottom of the cage as day 0 of pregnancy.The negative control group,low,medium and high dose group were exposed to neodymium nitrate(corresponding to ultra pure water,low,medium and high concentration of neodymium nitrate of 20,200 and 2000 mg/100 ml)for 21 days after delivery and feeding of young rats,and then the young rats were gavaged until 21 days after weaning,and then 1 male and 1 female were randomly selected for each litter,there were 16 rats in each dose group,half male and half female.The content of superoxide dismutase(SOD),malondialdehyde(MDA),catalase(CAT),glutathione peroxidase(GSH-Px),ascorbate peroxidase(APx),total antioxidant capacity(TAOC)and Ca^2+,calmodulin(CAM),brain-derived neurotrophic factor(BDNF)in the hippocampal tissue of young mice was determined by enzyme-linked immunosorbent assay.Results There was no significant difference in the relative weight of rat brain between different dose groups(P>0.05).There was statistically significant differences in the enzyme activities of CAT,MDA,GSH-PX and SOD in the hippocampal of female offspring mice at different concentrations(P<0.05),there was statistically significant differences in CAT,APX,GSH-PX,MDA and SOD in the hippocampal of male offspring mice at different concentrations(P<0.05).There was significant difference in the results of CAM measurement in hippocampus between male and female offspring in different dose groups(P<0.05),and there was significant difference in BDNF measurement in femal
