摘 要:为了建立饲料中25-羟基维生素D3的检测方法,采用高效液相色谱法通过空白样品添加回收试验验证检测方法的可靠性。样品用甲基叔丁基醚萃取,以甲醇∶水(V∶V=95∶5)为流动相,采用XSelectHSS T3色谱柱进行分离,紫外检测器TUV Detector(λ=265 nm)进行检测。结果表明:方法的线性范围为0.05—100μg∕mL,线性相关系数为0.9998,定量限(LOQ)为0.3 mg∕kg。添加水平在0.3 mg∕kg、0.6 mg∕kg和3 mg∕kg时,两种不同的饲料基质的回收率均在90%以上,RSD均在3.00%以下。该方法准确率高,操作简便,是饲料中25-羟基维生素D3较为理想的分析方法。The high performance liquid chromatography was applied to develop the method of analyzing 25-hydroxyvitamin D3 monohydrate in feed.Recovery test was performed to verify the reliability of the method.Samples were extracted with Methyl tertiary butyl ether(MTBE),then analyzed by separation on an XSelectHSS T3 chromatographic Column with methanol and H2O(V∶V=95∶5)solution as mobile phase,and detected with a TUV detector(λ=265 nm).The results showed that the linear range of 25-hydroxyvitamin D3 monohydrate was 0.05—100μg∕mL,and the linear correlation coefficient is 0.9998.The limit of quantitation(LOQ)was 0.3 mg∕kg.When 0.3 mg∕kg,0.6 mg∕kg or 3 mg∕kg 25-hydroxyvitamin D3 monohydrate was added to 3 kinds of feed,the average recovery rates were above 90%,and the relative standard deviations(RSD)were less than 2.9%.The method is accurate and simple operation,which is an ideal method for the analysis of 25-hydroxyvitamin D3 monohydrate in feed.
