摘  要:目的:探讨莪术提取物介导Wnt/β-catenin信号通路对肝癌Hep-G2细胞的影响。方法:将浓度为20、50、100μg/mL的β-榄香烯培养液加入Hep-G2细胞中,分别设为20μg/mL组、50μg/mL组和100μg/mL组,以不加β-榄香烯培养液作为对照组,RT-PCR、免疫印迹检测Hep-G2细胞Wnt1、β-catenin mRNA和蛋白水平,采用Transwell法、MTT法检测Hep-G2细胞的迁移和增殖情况,流式细胞仪检测Hep-G2细胞凋亡率。结果:对照组Wnt1、β-catenin mRNA水平均显著高于其他3个治疗组,100μg/mL组Wnt1、β-catenin mRNA水平显著低于20μg/mL组、50μg/mL组。对照组Wnt1、β-catenin蛋白表达高于其他3个治疗组,100μg/mL组Wnt1、β-catenin蛋白表达显著低于20μg/mL组、50μg/mL组。对照组Hep-G2细胞凋亡率显著低于其他3个治疗组,100μg/mL组Hep-G2细胞凋亡率显著高于20μg/mL组、50μg/mL组。100μg/mL组Hep-G2细胞迁移细胞数最低,显著低于其他3组。对照组Hep-G2细胞增殖能力显著高于其他3个治疗组,100μg/mL组Hep-G2细胞增殖能力显著低于20μg/mL组、50μg/mL组。结论:莪术提取物通过抑制Wnt/β-catenin信号通路活性,抑制肝癌细胞增殖和迁移。Objective:To explore the effects of zedoary turmeric extract on human hepatoma Hep-G2 cells through Wnt/β-elemeine signaling pathway.Methods:The human hepatoma Hep-G2 cells cultured in vitro were respectively treated with 0,20,50 and 100μg/mLβ-elemeine for 48 h.Then,Wnt1 andβ-catenin expressions in differently treated cells were detected using RT-PCR and Western blotting.Also,migration,proliferation,apoptosis of differently treated human Hep-G2 cells were examined.Results:The mRNA and protein levels of Wnt1 andβ-catenin in Hep-G2 cells treated with 0μg/mLβ-elemeine were statistically higher than those in cells treated with three concentrationsβ-elemeine.mRNA and protein levels of Wnt1 andβ-catenin in cells treated with 100μg/mLβ-elemeine were significantly lower than those in cells treated with 20 and 50μg/mLβ-elemeine.Usage ofβ-elemeine resulted in increased apoptotic cells in dose dependent manner.Similarly,Hep-G2 cells cultured withβ-elemeine exhibited statistical decrease in both of migration and proliferation,and these decreases were gradually enhanced along with increased dosage ofβ-elemeine.Conclusion:The zedoary turmeric extractβ-elemeine should inhibit proliferation and migration of human hepatoma Hep-G2 cells through suppressing Wnt/β-catenin signaling pathway.
