摘 要:目的通过响应面法优化超声波辅助提取黄精挥发油的工艺,并研究黄精挥发油的体外抗氧化活性。方法以提取溶剂、液料比、提取时间、提取温度为考察因素,在单因素实验的基础上,利用响应面法优化黄精挥发油的提取工艺。通过还原Fe3+能力和清除DPPH自由基的能力考察黄精挥发油的抗氧化活性。结果最佳提取溶剂为石油醚,提取工艺液料比为10:1,提取时间20 min,提取温度31℃,黄精中挥发油的得率2.035%。黄精挥发油还原Fe3+能力、清除DPPH自由基的能力与浓度呈正相关。结论优化的黄精挥发油提取工艺稳定,提取率高;黄精挥发油具有较强的抗氧化活性,可为进一步开发利用黄精药材提供依据。OBJECTIVE To optimize the process of ultrasonic-assisted extraction of volatile oil from Polygonatum sibiricum Red.by Response surface methodology and to study the antioxidant activity of the volatile oil of P.sibiricum.METHODS The extraction solvent,liquid to material ratio,extraction time and extraction temperature were taken as the investigation factors.On the basis of single factor experiment,the response surface method was used to optimize the extraction process of volatile oil from P.sibiricum.The antioxidant activity of the volatile oil of P.sibiricum was investigated by reducing the ability of Fe3+and scavenging DPPH free radicals.RESULTS By using the best extraction solvent petroleum ether,the best extraction process liquid to material ratio of 10:1,extraction time 20 min,and extraction temperature 31℃,the yield of volatile oil in PR was 2.035%.The ability of the volatile oil of P.sibiricum to reduce Fe3+and the ability to scavenge DPPH free radicals were positively correlated with the concentration.CONCLUSION The optimized extraction process of volatile oil from P.sibiricum is stable and the extraction rate is high.The volatile oil of PR has strong antioxidant activity,which provides a scientific basis for further development and utilization of P.sibiricum.
