摘 要:目的研究吴茱萸次碱对阿霉素致大鼠胚胎心肌细胞H9c2损伤的保护作用及其机制。方法选用1、5、10、20、40μmol·L^-1吴茱萸次碱作用H9c2心肌细胞48 h,用MTT法检测吴茱萸次碱对H9c2细胞的作用。选用1、5、10μmol·L^-1吴茱萸次碱或10μmol·L^-1右丙亚胺作为阳性对照预处理H9c2心肌细胞48 h,随后阿霉素(IC50=25μmol·L^-1)刺激4 h造成细胞损伤模型,采用MTT法、乳酸脱氢酶(LDH)释放法、流式细胞术、荧光探针DCFH-DA染色检测吴茱萸次碱预处理对阿霉素损伤H9c2心肌细胞的活力、完整性、凋亡及活性氧水平的影响。结果10μmol·L^-1以下吴茱萸次碱对H9c2心肌细胞无增殖抑制作用,而20μmol·L^-1以上吴茱萸次碱对H9c2细胞表现出增殖抑制作用。与阿霉素损伤组比较,5、10μmol·L^-1吴茱萸次碱预处理H9c2心肌细胞48 h后,可显著改善阿霉素损伤后H9c2细胞的活力,减少LDH的释放和细胞凋亡。与阿霉素损伤组比较,1、5、10μmol·L^-1吴茱萸次碱预处理组活性氧的水平均有降低。结论吴茱萸次碱对阿霉素引起H9c2心肌细胞的损伤具有保护作用,其机制可能与减少活性氧的释放有关。OBJECTIVE To study the protective effect and mechanism of rutaecarpine on Adriamycin-induced rat embryonic cardiomyocytes H9 c2 injury.METHODS 1,5,10,20,40μmol·L^-1 rutaecarpine were treated on H9 c2 myocardial cells for 48 h,and then the effect of rutaecarpine on H9 c2 cell were detected by MTT.After pretreatment of H9 c2 cells with 1,5,10μmol·L^-1 rutaecarpine as experimental group or 10μmol·L^-1 Dexrazoxane as positive control group for 48 hs,Adriamycin(IC50=25μmol·L^-1)was applied to stimulated the cells for 4 h.Then MTT,cytotoxicity LDH assay kit,flow cytometry and DCFH-DA probe were used to detect the effect of rutaecarpine on the survival rate,integrity,apoptosis and reactive oxygen species level.RESULTS Rutaecarpine below 10μmol·L^-1 had no proliferative inhibitory effect on H9 c2 cells,while above 20μmol·L^-1 showed inhibitory effect.Pretreatment of H9 c2 cells with 5,10μmol·L^-1 rutaecarpine for 48 h could significantly improve the activity of H9 c2 cells and reduced LDH release and apoptosis compared which Adriamycin injury group.Compared with Adriamycin injury group,the reactive oxygen species level of 1,5,10μmol·L^-1 rutaecarpine pretreatment group were reduced.CONCLUSION Rutaecarpine has protective effect on Adriamycin-induced H9 c2 myocardial cells injury,and the mechanism may be related to the reduction of reactive oxygen species release.
