摘  要:目的采用HPLC指纹图谱同时测定大花红景天中的红景天苷、酪醇和洛塞维。方法采用Cosmosil 5 C18-MS-Ⅱ色谱柱(250 mm×4.6 mm,5μm),流动相为0.1%甲酸溶液-乙腈,梯度洗脱,流速1.0 mL·min^-1,检测波长277 nm,建立了10批样品的指纹图谱;并测定样品中红景天苷、酪醇和洛塞维的含量。结果在10批样品的指纹图谱中确定了16个共有峰,样品相似度为0.942~0.991;与对照品比较,指认出红景天苷、酪醇和洛塞维成分;不同产地的10批大花红景天样品可分为3类;样品中红景天苷、酪醇和洛塞维的含量分别为0.69%~1.76%、0.16%~0.45%、0.07%~0.90%。结论所用方法快速实用、重复性好,在药材的质量控制方面有重要的价值。OBJECTIVE To simultaneously determine indicator components of salidroside,tyrosol and rosavin in Rhodiola crenulata by HPLC fingerprint.METHODS Cosmosil 5 C18-MS-Ⅱ column(250 mm×4.6 mm,5μm)was used as chromatographic column.0.1% aqueous formic acid and acetonitrile were used as mobile phase with gradient elution at a flow rate of 1.0 mL·min^-1.Detection wavelength was 277 nm.The fingerprints of 10 batches of samples were established.Contents of salidroside,tyrosol and rosavin in the samples were determined.RESULTS Sixteen common peaks were confirmed in the fingerprints of 10 batches of samples,and sample similarities were 0.942-0.991.After comparison with the reference substance,three components of salidroside,tyrosol and rosavin were identified.10 batches of R.crenulata samples from different regions were divided into 3 categories.The contents of salidroside,tyrosol and rosavin in the samples were 0.69%-1.76%,0.16%-0.45% and 0.07%-0.90%,respectively.CONCLUSION The method is fast and practical,has good repeatability,and has important application value in the quality control of medicinal materials.
