摘 要:为分析水貂杯状病毒中国分离株的遗传特征,本研究于2018年在山东地区某水貂养殖场内感染圆环病毒死亡的水貂中分离到1株水貂杯状病毒(mink calicivirus,MCV),命名为SD1株,本研究对其进行了PCR鉴定、电镜观察和遗传演化分析。结果显示,SD1株可在MDCK细胞上产生明显细胞病变。电镜观察可见病毒粒子表面成楔形杯状样凹陷,无囊膜,具有杯状病毒的形态特征。遗传演化分析显示,SD1株与2007年国内2株MCV的同源性分别为92.0%和91.2%,为水疮性病毒属新成员;RdRp基因编码的氨基酸序列比对分析显示其相对保守,蛋白变异不大,而vp1基因推导的氨基酸序列比对分析显示其变异较大,与国内分离株(MF677852)相比存在47处替换,该变异是否会引起病毒致病力的改变有待进一步解析。本研究为MCV流行株病原学研究提供参考,同时提示需重视MCV在水貂病毒感染中的作用。In 2018,a mink calicivirus(MCV)named with SD1,was isolated from a dead mink infected by the circovirus that was from a farm of Shandong province.The PCR identification,electron microscopic observation and whole genome analysis were carried out.MCV SD1 could produce significantly cytopathic effect on MDCK cells.Electron microscopic observation showed that the surface was cuneiform cup-like depression,and there was no envelope,both of which showed the morphological characteristics of calicivirus particles.Genetic evolution analysis showed that the homology between strain SD1 and two domestic strains was 92.0% and 91.2%,respectively,which belongs to the member of the genus vesivirus.The amino acid sequence alignment analysis of RdRp gene showed that it is relatively conservative,but the amino acid sequence alignment analysis of vp1 gene showed that vp1 protein sequences are variable.Compared with domestic isolates(MF677852),there are a totol of 47 substitutes in the vp1,which increase the degree of variation of SD1 strain.Whether these changes will increase viral pathogenicity needs to be further analyzed.This study can provide a reference for the etiological study of MCV epidemic strains in Shandong Province and suggests that more attention should be paid to the role of MCV in causing the mink infection.
