摘 要:分别在精液冷冻稀释液中添加不同浓度(0,25,50,75,100μmol/L)的金雀异黄酮,按照不同时间(30,60,90 min)孵育后冷冻。解冻后检测精子活力、顶体完整率和质膜完整性等表观指标;分光光度计检测各组Ca^2+浓度,金霉素染色检测精子获能状态。结果显示,当精液冷冻液中添加100μmol/L的金雀异黄酮孵育60 min后冷冻,解冻后精子活力提高,与对照组相比差异极显著(P<0.01),顶体完整率和质膜完整性略高于空白对照组;Ca^2+浓度低于其他处理组;B型精子数降低,与对照组相比差异显著(P<0.05)。综上所述,金雀异黄酮能够通过抑制精子冷冻过程中的被动获能,显著改善猪精液的冻后质量。Different concentrations(0,25,50,75 and 100μmol/L)of genistein were added to the semen cryopreservation solution and frozen after incubation at different times(30,60 and 90 min).After thawing,the apparent indicators such as sperm motility,acrosome integrity and plasma membrane integrity were detected.The concentration of Ca^2+ in each group was detected by spectrophotometer,and the sperm capacitation status was detected by chlortetracycline staining.The results showed that when the supernatant was incubated with 100μmol/L of genistein for 60 min,the sperm motility increased after thawing,which was significantly different from the control group(P<0.01).Acrosome integrity rate and plasma membrane integrity were slightly higher than the control group.And Ca^2+ concentration was lower than other treatment groups.Compared with the control group,B-type sperm count decreased significantly(P<0.05).In conclusion,genistein can significantly improve the post-freezing quality of boar semen by inhibiting passive capacitation during sperm freezing.
