摘  要:采用羟基磷灰石(HAp)纳米粒子为稳定粒子,海藻酸钠和丙烯酰胺溶液为水相,聚乳酸-羟基乙酸共聚物(PLGA)二氯甲烷溶液为油相,制备水包油(O/W)型Pickering乳液,通过使水相中的海藻酸钠发生交联和引发丙烯酰胺单体聚合,制备得到载PLGA微球的聚丙烯酰胺-海藻酸钠双网络多孔凝胶支架,并采用万能力学机测试了支架的拉伸性能,使用扫描电子显微镜(SEM)表征了支架的结构。另外探讨了装载抗菌药物恩诺沙星支架的药物释放性能,并评价了载药支架的抗菌性能。研究结果表明:双网络凝胶支架具备优良的拉伸性能,当海藻酸钠为1%、羟基磷灰石为3%、丙烯酰胺为4.5mol/L和交联剂为0.3mL时,可制得拉伸性能最好的水凝胶支架。多孔支架内含有微球,适量的交联剂能使多孔支架结构更为完整,过多的交联剂会导致孔的均匀性和通透性变差。乳液制备条件的改变对微球的大小没有明显影响。载恩诺沙星的多孔凝胶支架具有良好的药物缓释能力,并且对大肠杆菌和金黄色葡萄球菌有很好的抑菌效果,说明该支架可以很好地装载疏水性药物。The Pickering emulsion was firstly prepared using sodium alginate and acrylamide as the aqueous phase and the dichloromethane solution of poly(lactic-co-glycolic acid)(PLGA)as the oil phase,and at the same time hydroxyapatite(HAp)worked as the stable particles in the emulsion.Taking advantages of the crosslinking reaction of sodium alginate and polymerization reaction of acrylamide in the Pickering emulsion PLGA microspheres loaded polyacrylamide-sodium alginate porous gel scaffolds with double networks were then formed.Using the control variable method,the tensile performance test of the gel scaffolds prepared under the different preparation condition had been performed.The porous structure of the gel scaffolds was characterized by scanning electron microscope(SEM).In addition,the drug release performance of antibacterial drug enrofloxacin loaded porous gel scaffolds was discussed,and the antibacterial properties of the enrofloxacin loaded porous gel scaffolds was evaluated by disc diffusion method.The experimental results indicate that the double network gel prepared with 1%sodium alginate,3%hydroxyapatite,4.5 mol/L acrylamide and 0.3 mL crosslinking agent exhibits the best tensile properties.And proper amount of crosslinking agent can make the porous scaffolds structure complete,but superfluous crosslinking agent can make the hole uniformity and permeability decline.The emulsion preparation condition change have no significant effect on the size of the microspheres.The drug-loaded porous gel scaffolds show good drug slow release capacity,and excellent inhibition effect against Escherichia coli and Staphylococcus aureus,suggesting that the gel scaffold can be well loaded with hydrophobic drugs.
