摘 要:为了解腱蛋白样蛋白1(Chordin-Like1,CHRDL1)基因的表达量与蛋壳颜色深浅之间的规律性,本实验以赤水乌骨鸡为研究对象,挑选180、360、540日龄母鸡各100只,利用双重PCR对其基因分型,再根据基因型(纯合型、杂合型)和蛋壳颜色(深绿色、浅绿色、淡绿色)进行分类。采集360日龄赤水乌骨鸡的蛋壳腺进行转录组测序分析,并利用实时荧光定量PCR检测CHRDL1基因在不同产蛋期赤水乌骨鸡蛋壳腺和肝脏中的表达量,验证转录组测序分析的结果。结果显示:CHRDL1基因的表达量与蛋壳中胆绿素含量呈极显著负相关,相关系数为-0.8883,且随着蛋壳绿色的加深而降低,验证了前期转录组测序显示CHRDL1基因为蛋壳绿色的负调控基因的结果。这一研究结果可以为绿壳性状的遗传机制研究提供基础资料。To study the relationship between CHRDL1 gene expression and eggshell color depth,in this study,selected different laying period of 180 days of age,360 days of age,540 days of age of Chishui Silky Fowl hens as experimental animals.Duplex PCR method was used to divided them into different types.Then according to the genotypes(homozygous,heterozygous)and shell color(dark green,green and light green)were then classified.The eggshell glands of 360-day-old Chishui silky fowl chicken were collected for transcriptome sequencing analysis.Three hens were selected from each types of each period randomly,gathering the shell glands and livers.Total RNA was extracted and reversely transcribed into cDNA,then The expressions of SPIA1 gene in different tissue were detected by Quantitative Real-time PCR method.The expression level of CHRDL1 gene was significantly negatively correlated with the content of bilirutin in eggshell,with a correlation coefficient of -0.8883,which decreased with the deepening of eggshell green.This verified the results of early transcriptome sequencing showing that CHRDL1 gene was a negative regulatory gene in eggshell green.The results of this study could provide basic information for the study of genetic mechanism of green shell traits.
