摘 要:氮是水稻生产发育过程中所必须的大量营养元素,但是大量的氮肥投入在提高产量的同时对环境造成了严重危害。选用含有氮高效基因的水稻品种提高水稻自身的氮肥利用效率是减少氮肥使用量降低环境氮污染的最有效途径之一。NRT1.1B是一个影响水稻籼、粳亚种间氮肥利用效率的关键基因,主要分布在籼稻品种中。为了筛选携带NRT1.1B基因的粳稻资源,根据氮高效基因NRT1.1B与其等位基因nrt1.1b在功能区域存在的单核苷酸变异,设计和筛选出NRT1.1B的等位基因特异PCR功能标记1nrt/1NRT。结合测序分析验证,1nrt/1NRT可以准确快速鉴定出NRT1.1B的不同基因型。利用1nrt/1NRT对71份籼稻品种和134份粳稻品种进行NRT1.1B基因型检测,结果表明71份籼稻品种均携带NRT1.1B基因,134份粳稻品种均携带nrt1.1b基因。进一步对172太湖流域地方粳稻资源和99份粳稻育种中间品系进行NRT1.1B基因型检测,结果发现粳稻品系‘常粳144’携带NRT1.1B基因,测序分析也进一步证实了该结果。本研究为利用NRT1.1B改良粳稻氮高效育种提供了科学依据。Nitrogen is a fundamental macronutrient required for plant growth and development in rice,but a large amount of nitrogen input not only increases the yield,but also causes serious harm to the environment.Development and application of varieties carrying high nitrogen use efficient alleles is one of the most effective strategies to reduce agricultural nitrogen pollution.NRT1.1 B is a key gene for nitrogen use efficiency diverge between indica and japonica rice.However,most of the NRT1.1 B variations enhancing nitrogen use exist in indica rice.In order to screen japonica genetic resources with beneficial NRT1.1 B alleles,a molecular marker,1 nrt/1 NRT,was developed based on the single nucleotide polymorphism in the functional region between NRT1.1 B and nrt1.1 b alleles.The different genotypes of NRT1.1 B can be accurately distinguished by using the 1 nrt/1 NRT marker.The NRT1.1 B variations in 71 indica varieties and 134 japonica varieties were identified.We found that all the indica rice carry NRT1.1 B,while all the japonica rice carry nrt1.1 b.Moreover,172 local japonica rice at Taihu Lake area and 99 japonica breeding lines were genotyped by the 1 nrt/1 NRT marker.Fortunately,we found that’Changjing144’,a breeding line,carries the NRT1.1 B allele.This result was further confirmed by DNA sequencing.Our data literately facilitates the use of NRT1.1 B gene in improving nitrogen use efficiency for japonica rice breeing.
