摘 要:建立雪上一枝蒿中雪上一枝蒿甲素的含量测定方法,并考察不同产地、不同炮制方式对雪上一枝蒿甲素含量的影响。使用Waters Xterra MS C 18柱(5μm,4.6 mm×250 mm),以磷酸盐缓溶液和甲醇为流动相,进行梯度洗脱;流速:1.0 mL·min^-1,检测波长:210 nm。结果显示,雪上一枝蒿甲素在0.0515~10.3000μg范围内呈良好的线性关系(r=0.9999),回收率为93.57%~100.55%(n=9),RSD值为3.0%。本法准确、可靠,重现性好,适用于测定雪上一枝蒿中雪上一枝蒿甲素含量;不同产地不同批次药材中的甲素含量存在显著差异;炮制方式对雪上一枝蒿甲素含量无显著影响。A method for the determination of bullatine A content in Aconitum brachypodum Diels was established,and the influence of different geographica areas and different processing methods on the content of bullatine A in Aconitum brachypodum Diels.was investigated.The content of derivatives was determined by HPLC.HPLC was performed on Waters Xterra C 18 column(5μm,4.6 mm×250 mm),phosphate buffer solution and methanol as mobile phase,using gradient elution with flow rate of 1.0 mL·min-1,and detection wavelength of 210 nm.Bullatine A had a good linear relationship in the range of 0.0515~10.3000μg(r=0.9999),the recovery rate was 93.57%~100.55%(n=9),and RSD value was 3.0%.This method was accurate,reliable and reproducible.It was suitable for the determination of the artemisinin content of Artemisia sphaerocephala.There were significant differences in the content of artemisinin in different batches of medicinal materials from different origins.Different processing methods had no significant effect on the content of bullatine A.
