摘 要:以继代阶段的多花黄精组培苗为试验材料,比较了卡拉胶为凝固剂的常规培养基、滤纸、纱布为固着物的瓶内浅层液体培养基和沙子为固着物的培养基的生根效果,并比较了多花黄精组培生根块茎的不同冷藏时间和不同移栽深度对块茎出苗时间、整齐度和成活率等方面的影响。结果表明,以沙子为固着物的培养基最佳,根系粗壮,无需洗苗,冷藏70~80 d能大大缩短块茎的休眠时间,加快块茎的出苗速度。种植深度以基质表层为好,可大大减少块茎腐烂的机率,提高多花黄精组培苗块茎的成活率。Tissue cultured plants of Polygonatum cyrtonema were cultured on different rooting mediums to compare the rooting effects of conventional medium with carrageenan as coagulant,shallow liquid medium with filter paper as fixative,shallow liquid medium with gauze as fixative and medium with sand as fixative,and the effects of different cold storage time and transplanting depths on the emergence time,uniformity and survival rate of rooted tissue culture tubers of P.cyrtonema were compared.The results showed that the sand is the best fixative in the rooting medium,on which tissue culture plants produced thick and strong roots and were not necessarily rinsed.Cold storage for 70-80 days shortened the dormancy time of tissue cultured tubers and speeded up the plant emergence from the tissue cultured tubers.It is better to plant the tissue culture tubers on the surface layer of the substrate,which could greatly reduce possible tuber decay and improve the survival rate of tissue culture plants.
