摘 要:红叶石楠为新兴彩叶树种,其叶色会随季节而变,具较高的观赏价值,但其叶色转变的调控机制尚未见报道.本研究拟探析红叶石楠叶色转变期的基因组DNA甲基化变异,进而为解析红叶石楠叶色转变的调控机理提供新思路.本研究于红叶石楠叶色转变期分别收取红色、褐色、绿色发育相同的幼嫩叶片为材料,通过甲基化敏感扩增多态性(MSAP)技术分析其基因组DNA甲基化的变异情况.结果表明,以筛选出的20对引物组合,在3种叶片材料中分别扩增产生325、282和257个清晰可辨的基因位点,发生甲基化的位点分别占49.23%、70.57%和87.94%,表明红叶石楠叶色由红转绿过程中,其基因组DNA甲基化水平显著提升.红叶石楠叶色由红转绿的渐变过程中,其基因组甲基化比例呈现递增的趋势,推测其基因组甲基化变异参与调控红叶石楠的叶色转变.本研究为进一步探究红叶石楠叶色转变的表观遗传调控机理奠定基础.Photinia frasery is an emerging color-leafed tree with high ornamental value,and its leaf color changes with the season.However,the regulatory mechanism of the leaf color transition is unclear.This study intends to analyze the genomic DNA methylation variation in the leaf color transition period of Photinia fras⁃ery,thus to provide new ideas for the analysis of the regulation mechanism of the leaf color transition of Pho⁃tinia frasery.In this study,during the leaf color transition period of Photinia frasery,young leaves with the same development time in red,brown,and green were collected as materials.The genomic DNA methylation was analyzed by methylation-sensitive amplification polymorphism(MSAP).And the results showed that 325,282,and 257 clearly identified gene loci were amplified in the three leaf materials of red,brown,and green by using the selected 20 primer combinations,respectively.The methylation sites accounted for 49.23%,70.57%,and 87.94%,indicating that the genomic DNA methylation levels were significantly induced during the leaf color transition from red to green.In the gradual process of the leaf color change of Photinia frasery,its genomic methylation ratio showed an increasing trend.It is speculated that the genomic methylation varia⁃tion is involved in regulating the leaf color change of Photinia frasery.This study lays a foundation for further exploring the epigenetic regulation mechanism of leaf color transition of Photinia frasery.
