摘  要:目的:探讨艾灸对类风湿性关节炎的抗炎效应及对可溶性程序性细胞死亡因子-1(soluble Programmed Death-1,sPD-1)水平的调节作用。方法:选用类风湿性关节炎(Rheumatoid Arthritis,RA)大鼠和日本大耳白兔为研究对象,采用弗氏完全佐剂造模,艾灸“肾俞”“足三里”治疗,治疗结束采集大鼠血清、家兔滑膜液,用酶联免疫法检测滑膜液和血清sPD-1水平,滑膜液和血清干扰素γ(Interferon-γ,IFN-γ)表达。结果:与空白对照组相比较,模型组滑膜液和血清中的sPD-1和IFN-γ水平均明显升高(P<0.01);与模型组比相较,艾灸组滑膜液的sPD-1和IFN-γ水平均明显下降(P<0.05),血清中sPD-1和IFN-γ水平均显著降低(P<0.01)。结论:艾灸可以降低RA模型中异常升高的sPD-1和IFN-γ水平,艾灸可能通过抑制sPD-1表达而减低炎症反应,sPD-1可能是艾灸抗炎效应的重要环节。Objective:To investigate the anti-inflammatory effect of moxibustion on rheumatoid arthritis(RA)and the regulation of soluble programmed death-1(sPD-1)level.Methods:Sprague Dawley rats and Japanese white rabbits were selected as the research objects.The models were made with Freund’s complete adjuvant and treated with moxibustion at“Shenshu”(BL23)and“Zusanli”(ST36).At the end of the treatment,the serumof rats and synovial fluid of rabbits were collected.The content of sPD-1 in synovial fluid and serum and the expression of IFN-γin synovial fluid and serum were detected by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the blank groups,the content of sPD-1 in rabbits’synovial fluid and rats’serum in model groups were significantly increased(P<0.01);compared with model groups,the content of sPD-1 in rabbits’synovial fluid and rats’serum in moxibustion groups were significantly decreased(P<0.05).Compared with the blank groups,the content of IFN-γin synovial fluid and serum in the model groups were significantly increased(P<0.01),and the content of IFN-γin synovial fluid and serum of moxibustion groups were significantly lower than that of model groups(P<0.05,P<0.01).Conclusion:Moxibustion can reduce the abnormally increased contents of sPD-1 and IFN-γin RA models.Moxibustion may reduce inflammatory response by inhibiting the expression of sPD-1.sPD-1 may be an important link in the anti-inflammatory effect of moxibustion.
