摘  要:目的探讨孕激素对白假丝酵母菌刺激的小鼠腹腔巨噬细胞NLRP3炎性体表达的影响。方法体外培养小鼠腹腔巨噬细胞,将其分为5组:空白对照组、白假丝酵母菌组、白假丝酵母菌+10^-9mol/L孕酮组、白假丝酵母菌+10^-8mol/L孕酮组、白假丝酵母菌+10^-7mol/L孕酮组。利用实时荧光定量PCR检测细胞内NLRP3基因表达情况,利用Western blot检测细胞内NLRP3、Caspase-1 p20和NF-κB p65的表达变化,利用酶联免疫吸附试验法检测细胞培养上清中白细胞介素-1β(IL-1β)和白细胞介素-18(IL-18)的含量。结果白假丝酵母菌刺激小鼠腹腔巨噬细胞后引起细胞内NLRP3和Caspase-1 p20蛋白表达水平升高,NF-κB活性增强,细胞培养上清中的IL-1β和IL-18含量明显升高,白假丝酵母菌组和空白对照组比较差异均有统计学意义(均P<0.01);加入孕酮预处理后,随着孕酮作用浓度的增高,细胞内NLRP3、Caspase-1 p20蛋白表达水平和NF-κB活性明显降低,细胞培养上清中IL-1β和IL-18含量亦明显降低,和白假丝酵母菌组比较,差异均有统计学意义(均P<0.01)。结论孕酮可抑制白假丝酵母菌刺激引起的巨噬细胞内NLRP3表达及Caspase-1活化和IL-1β、IL-18的分泌,且这种作用可能是通过抑制NF-κB活性实现的。Objective To explore the effects of progesterone on the expression of NLRP3 inflammasome in murine peritoneal macrophages stimulated with Candida albicans(C.albicans).Methods Murine peritoneal macrophages were cultured in vitro and divided into five groups:blank group,C.albicans group,C.albicans+10^-9 mol/L progesterone group,C.albicans+10^-8mol/L progesterone group,and C.albicans+10^-7mol/L progesterone group.The mRNA expression of NLRP3 in macrophages was detected by real-time PCR.The protein expression levels of NLRP3,Caspase-1 p20 and NF-κB p65 in macrophages were determined by Western blotting.The levels of interleukin-1β(IL-1β)and interleukin-18(IL-18)in the culture supernatant were measured by enzyme-linked immunosorbent assay(ELISA).Results The expression levels of NLRP3 and Caspase-1 p20,the activity of NF-κB in murine peritoneal macrophages stimulated with C.albicans,and the levels of IL-1βand IL-18 in the supernatant were increased significantly than those in the controls(P<0.01).After pretreatment with progesterone,the expression levels of NLRP3 and Caspase-1 p20,the activity of NF-κB in macrophages,and the levels of IL-1βand IL-18 in the supernatant were decreased with the increasing of progesterone concentrations.Conclusion Progesterone can dose-dependently inhibit the expression of NLRP3,the activation of Caspase-1 and the secretion of pro-inflammatory cytokines IL-1βand IL-18 in macrophages stimulated with C.albicans.And the inhibitory role of progesterone may be achieved by inhibiting NF-κB activity.
