摘　　要：目的探究过表达微小RNA-200c(miR-200c)对表皮葡萄球菌感染所致的腹膜透析(PD)相关腹膜炎大鼠的干预效果及相关作用机制。方法选取40只SD健康雄性大鼠,10只为正常组,其余30只建立表皮葡萄球菌感染所致PD相关腹膜炎大鼠模型,并分为模型组、抑制miR-200c组、过表达miR-200c组,正常组、模型组使用生理盐水干预,抑制miR-200c组大鼠使用antagomiR-200c进行干预,过表达miR-200c组大鼠使用agomiR-200c进行干预。对各组大鼠腹膜功能、腹水白细胞计数、菌落计数进行统计,酶联免疫吸附实验法检测白细胞介素-8(IL-8)、白细胞介素-17(IL-17)水平,免疫透射比浊法检测免疫球蛋白水平,蛋白免疫印迹法检测ZEB1、ZEB2、E-钙黏蛋白表达。结果过表达miR-200c组大鼠Durea/Purea、葡萄糖转运率、菌落计数低于模型组、抑制miR-200c组,高于正常组;超滤量低于正常组,高于模型组、抑制miR-200c组(P<0.05)。过表达miR-200c组大鼠IL-8、IL-17、IgA、IgM、IgG水平、ZEB1、ZEB2相对表达量均低于模型组、抑制miR-200c组,高于正常组;E-钙黏蛋白表达低于正常组,高于模型组、抑制miR-200c组(P<0.05)。结论过表达miR-200c对表皮葡萄球菌感染所致PD相关腹膜炎大鼠进行干预,能够提升大鼠腹膜功能,改善腹水白细胞计数及表皮葡萄球菌计数,减轻大鼠炎症反应,提升大鼠免疫功能,调控ZEB1、ZEB2、E-钙黏蛋白表达,具有一定的腹膜保护作用。OBJECTIVE To explore the effect of over expressed microRNA-200 c(miR-200 c)on intervention to peritoneal dialysis(PD)related peritonitis induced by Staphylococcus epidermidis and its mechanisms.METHODS Totally 40 SD healthy male rats were selected,10 rats were assigned as the normal group,the other 30 rats were established as PD related peritonitis model induced by S.epidermidis infection and were divided into the model group,the inhibition of miR-200 c group,the overexpression of miR-200 c group.The normal group and the model group were intervened with normal saline,the inhibition of miR-200 c group was treated with antiagor-200 c,and the overexpression of miR-200 c group was treated with agomiR-200 c.The peritoneal function,leukocyte counts in ascites and colony counts of the three groups were analyzed,the levels of interleukin-8(IL-8)and interleukin-17(IL-17)were detected by using enzyme-linked immunosorbent assay,the levels of immunoglobulins were detected with the use of immunotransmission turbidimetry,and the expressions of ZEB1,ZEB2 and E-cadherin were detected by Western blot.RESULTS Durea/Pura,glucose transport rate and colony counts were significantly lower in the overexpression miR-200 c group than in the model group and the inhibition miR-200 c group,significantly higher in the overexpression miR-200 c group than in the normal group;the ultrafiltration volume was significantly lower in the overexpression miR-200 c group than in the normal group,significantly higher in the overexpression miR-200 c group than in the model group and the inhibition miR-200 c group(P<0.05).The levels of IL-8,IL-17,IgA,IgM,IgG,relative expression of ZEB1 and ZEB2 were significantly lower in the over expression miR-200 c group than in the model group and the inhibition miR-200 c group,and the levels of above indexes were significantly higher in the over expression miR-200 c group than in the normal group;the expression of E-cadherin was significantly higher in the over expression miR-200 c group than in the model group and t
